Abstract


Shigella isolated from chicken can cause chicken dysentery and blood stool, if illeness is serious, it can lead to chicken’s death. As a new infectious disease, chicken shigellosis are always misdiagnosed to be chicken white diarrhea or chicken coccidiosis. Untile now, there are lack of enough understanding of how Shigella isolated from chicken cause infectious and its pathogenic mechanism in vivo, which influence prevention and control of the disease. so, we need to have a deep understanding of pathogenic process and mechanism of Shigella isolated from chicken. Studying bacterium’s deynamic distribution in animal’s vivo is the best way to understand the disease occurrence and development process of the best method. So, to master the pathogenic process of Shigella isolated from chicken requires to firstly know the bacteria’s dynamic distribution in vivo.In this experiment, a PCR detection method were established firstly, secondly experimental chicken were infected artificially by challenging Shigella isolated from chicken to make a pathological model of chicken shigellosis. In vivo the bacteria’s dynamic distribution of Shigella isolated in chicken were studied by using bacteria separation and identification method and PCR detection, revealing the occurrence, development process and pathogenesis mechanism of chicken shigellosis, providing the theory basis for epidemiology investigation, diagnosis and prevention.The results were as follows:1. In this experiment, specifice to Shigella invasive antigen gene ipaH ,we established a PCR detection system for Shigella isolated from chicken.The results showed that this PCR detection system has high specifity and sensitivity, the limit of detection is 126 cfu/mL. If the PCR detection system were used to detect Shigella isolated from chicken in clinical samples, detection time will be shorten, detection accuracy rate will be improved, providing technical means for prevention and control of chicken shigellosis .2. According to the detection results of dynamic distribution of Shigella isolated from chicken in infected chicken vivo, Shigella isolated from chicken can be detected in heart blood, liver, spleen, lungs, kidney and duodenum, the detection results of Shigella isolated from chicken in viscera organs are different at different time after bacteria challenge. Shigella isolated from chicken can not be dectected in chicken vivo in seventh day. From the whole bacterium distribution, the course of chicken shigellosis reachs to the peak of infection, later, go health gradually. Liver, spleen and kidney are major colonization organ of Shigella isolated from chicken, the distribution of Shigella isolated from chicken have certain individual differences in heart, lungs and duodenum. In the later diagnostic work, collecting the liver, spleen and kidney for detecting Shigella isolated from chicken has more pertinency, so, can reduce the workload.3. According to detection results of Shigella isolated from chicken in chicken rectum content, at 6 h after infection, Shigella isolated from chicken can be detected up to the second day. Chicken excreting bacterium has individual difference, Shigella isolated from chicken can be detected in several chickens, the detection rate is very low. In addition, live Shigella isolated from chicken can be excreted to external environment via feces contaminating environment, healthy chicken could be infected via digestive tract.4. Comparaing distribution that Shigella isolated from chicken in chicken with bacteria separation method and distribution achieved with PCR detection, we can see that: detecting Shigella isolated from chicken in samples with traditional culture method and PCR, detectablerate will be higher than that with bacteria separation method only. In clinical, using PCR detection method to detect whether disease material culture has Shigella isolated from chicken can save diagnosis time, providing guarantee for timely treat.
