Significance of IL-4 and IL-12 Expressions in the Cervical Tumor and Relation to Taxol Allergy in Cervical Cancer

Abstract


Objective:To examine the expression levels of interleukin (IL)-4 and IL-12 in the tissues from normal cervix, cervical intraepithelial neoplasia (CIN) I group, CINⅡ-Ⅲgroup, and cervical cancer group respectively, and to investigate the relationships between the expressions of ILs and the development and progression of the cervical disease. Furthermore, the possible involvement of IL-4 andⅡ-12 in the Taxol allergy in the patients with cervical cancer who underwent chemotherapy based on Taxol was also studied.Methods:Total RNA was extracted from paraffin-embedded tissues which were obatained from the patients with CINⅠ(15 cases), CINⅡ-Ⅲ(15 cases) and cervical cancer (30 cases). The cervical cancer patients who underwent taxol-based chemotherapy were divided into taxol allery group (9 cases) and non-allergy group (21 cases). The normal cervical tissues which were obtained from the patients with benign diseases were used as control (15 cases). Reverse Transcription Polymcrase Chain Reaction (RT-PCR) was performed and expressions of IL-4、IL-12p35 and IL-12p40 were analyzed. Whether the IL-4 and IL-12 were involved in the taxol allergic process was also investigated.Results:1. The expression of IL-4 mRNA in the cervical cancer was higher than those of tissues from normal cervix, CIN I and CINII-III group. The IL-4 mRNA expression in CINⅡ-Ⅲgroup was significantly higher than those of normal cervical tissues and CIN I groups. On the contrary, there was no significant difference between the normal cervical tissues and CIN I group (P>0.05).2. The expressions of IL-12p35 mRNA and IL-12p40 mRNA in the cervical cancer were higher than those of normal cervical tissues and CIN groups. In the precancerous group (CINⅡand CINⅢ), the IL-12 mRNA expression was significantly higher than that of normal cervix and CINⅠgroup. The expression of IL-12p35 mRNA in the CINII-Ⅲgroup was significantly higher, compared to CINⅠgroup. There were significant differences among IL-12 in all the above groups (P<0.05). On the contrary, IL-12p40 mRNA did not show any significant difference between the CINⅠand CINⅡ-Ⅲgroup (P>0.05).3. The expression of IL-4 mRNA in Taxol allergy group of the cervical cancer was higher than non-allergy group. Rather, IL-12p35 mRNA and IL-12p40 mRNA were lower than non-allergy group. There were significant differences among all the above groups (P<0.05).Conclusions:1. The increased IL-4 expression and decreased IL-12 expression may promote the development and progression of the cervical diseases. These effects may be closely related to the loss of anti-tumor and immune functions of IL genes.2. The aberrant expressions of TL-4 and IL-12 may be involved in the taxol allergical process in the cervical cancer patients who underwent taxol-based chemotherapy. The increased IL-4 gene expression and decreased IL-12 expression levels may result in the imbalance of immune system function and increase the incidence of taxol allergy.
