Abstract


Objective Make the rabbit cerebral vasospasm model of subarachnoid hemorrhage. To study changes in rabbit models of behavior scores, the degree of cerebral vasospasm, expression of Ras-GTP and ERK1/2 phosphorylation, apoptosis of endothelial cells.Methods The rabbit cerebral vasospasm model of subarachnoid hemorrhage, with the method that injection blood into the cisterna magna twice. Experimental groups: the first large group was divided into control group, SAH group, SAH + DMSO group, SAH + FTI-277 group; the second largest group was divided into control group, SAH 3 days group, SAH 5 days group, SAH 7 days group.①Evaluation of behavioral changes in rabbit model;②Measure cross-sectional area of vascular changes by computer software;③Detect Phosphorylation of ERK1/2 levels by immunohistochemistry;④Detect expression Ras-GTP and phosphorylated ERK1/2 by WESTERN BLOT;⑤Detect apoptosisr of abbit cerebral vascular endothelial cell by TUNEL.Results①SAH group, SAH+DMSO group than SAH+FTI-277 group behavior score lower, statistically significant difference.②SAH group, SAH+DMSO group and SAH+FTI-277 group was statistically significant cerebral vascular cross-sectional area, SAH+FTI-277 group and control group was not statistically significant.③SAH group and SAH+DMSO group deeper immunohistochemistry (phosphorylation of ERK1 / 2 levels higher), SAH + FTI-277 group and control group phosphorylation of ERK1 / 2 levels lower.④the second large group, SAH group and SAH+DMSO group, Ras-GTP and phosphorylated ERK1/2 levels increase significantly, are different from the control group and SAH + FTI-277 group; the second largest group, SAH 3 days group and SAH 5 days group’s Ras-GTP and phosphorylated ERK1 / 2 levels increase, SAH 7 days group’s Ras-GTP and phosphorylated ERK1 / 2 levels decreased.⑤SAH group and SAH + DMSO group more cerebral vascular endothelial cell apoptosis. Control group and SAH + FTI-277 rare apoptotic cells.Conclusion Production of cerebral vasospasm after subarachnoid hemorrhage rabbit model is satisfactory in the experiment. After SAH, Ras-GTP and phosphorylation ERK1/2 both increase significantly. The activation levels in time with the pathophysiology of cerebral vasospasm corresponding changes. This suggests that Ras in the pathologic process plays an important role.
