ObjectiveTo construct a lentiviral vector with c-myc gene, including wide type and mutation type, and provide research tools to investigate the regulation of c-myc gene in cell cycle and apoptosis.MethodsThe lentiviral vector with GFP was constructed by PCR and gateway technology and identified by PCR and gene sequencing. Transfect into the package cell 293FT with pLV/helper-SL3、pLV/helper-SL4 and pLV/helper-SL5 by lipofectin to produce mature lentivirus. Measure the virus titer. Infect HCC1937 to defect its activity.ResultsAccording to PCR and gene sequencing, successfully construct the myc-lentiviral vectors, the virus titer results were 6.1×107 TU/ml and 5.65×107 TU/ml, respectively.ConclusionSuccessfully construct the myc-lentiviral vector and package efficient lentivirus particles. Lay the material foundation and pave the way for future study of the signal transduction pathway of cell cycle and apoptosis by c-myc gene.
