Abstract


Siniperca chuatsi is the most important economic Mandarin fish because of the fast growth. In recent years, due to the survival environment destruction and artificial selection, the germplasm resources of Siniperca chuatsi have degenerated continually. Therefore, the collection and conservation of the germplasm resources of Siniperca chuatsi, analysis of the genetic diversity and the the genetic variation between groups are useful for the protection, monitoring and breeding improvment of germplasm resources of Siniperca at the molecular level.1、TRAP was used for comparison of genetic diversity in both protospecies and cultured populations of Siniperca chuatsi.Twenty-one primer combinations selected from 30 primers combinations produced good amplified patterns and 330 amplified polymorphic loci from two populations were obtained. The numbers of polymorphic loci in protospecies and cultured populations were 149 and 181 respectively. In protospecies population, each combination average produced 8.62 a polymorphisms loci,the polymorphism information content (PIC) was 0.6339 while the corresponding values of the cultured population are 7.1 and 0.5811 respectively. The results indicated that, the genetic diversity of cultured population has slightly decreased relatively. With some primer combinations, there is a gene missing phenomenon. This will offer the potential molecular markers for degradation of germplasm resources of Siniperca chuatsi.2、In this study, twenty pairs of published micro satellite primers that isolated and designed by ourselves were used to detect the genetic diversity in both protospecies and cultured populations of Siniperca chuatsi. The results showed that there were 55.21% polymorphism loci in the protospecies population while 44.79% loci in the cultured population. We also found that the effective number of alleles, the Shannon index, the observed heterozygosity, the Nei’s expected heterozygosity and the polymorphism information content (PIC) of the protospecies population were 2.7390,1.0780,0.5708,0.5295 and 0.5353, respectively, while in the cultured population, their corresponding values are 2.1284,0.6889,0.3839,0.4011 and 0.5043 respectively. All above results suggested that the protospecies population remain higher relatively genetic diversity. Germplasm resources protection effect is good, and breeding population genome appear lost phenomenon, genetic diversity declines, which is consistent with the TRAP.
