Abstract


In this thesis, the ethylenediaminetetraacetic acid anhydride (EDTAD), butanediamine (BDA) and the weight average molecular weight of 500 amino-capped polyethylene glycol as raw materials, synthesized a novel amphoteric pH sensitive hydrogel, The hydrogel along with biodegradable, pH sensitivity and good biocompatibility. Using nuclear magnetic resonance (NMR), infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and chemical analysis methods to characterize the chemical structure of the material and the pH sensitivity, and initially examined the hydrolysis properties of the material. The following are the main contents and conclusions obtained: (1) to ethylenediaminetetraacetic acid anhydride (EDTAD) and butanediamine (BDA) for raw materials, ring-opening reaction of N-acylated by anhydride side chains containing only carboxyl a straight-chain copolymer PEB-COOH, and then with N-hydroxy succinimide (NHS) and dicyclohexyl carbodiimide (DCC) activation of the carboxyl group in the PEB-COOH, weight average molecular weight of 500 amino capped the polyethylene glycol (ATPEG500) as crosslinker, the monomer connection between all of the amide bond, the side chain carboxyl-containing only a crosslinked network polymer PEB-ATPEG500-COOH. (2) FTIR, NMR and SEM results showed that the synthetic method of this study in successful preparation of linear copolymers PEB-COOH, and the side chain carboxyl-containing only the PEB-ATPEG500-COOH crosslinked polymer networks. (3) a number average molecular weight of ninhydrin color method for the determination of the PEB-COOH. The test results show that the control process conditions, the PEB-COOH can be prepared by different molecular weights. (4) pH sensitivity tests show that the sensitive range of three pH the PEB-ATPEG500-COOH, pH 2-4, the pH 6-7 and pH 10-11, showed significant gender pH sensitivity; With the cross-linking The dosage increase the sensitivity of the pH of the hydrogel gradually weakened. (5) were examined PEB-ATPEG500-COOH 37oC under conditions ph2, 7,11 medium swelling kinetics. The results show that the swelling process of hydrogels PEB-ATPEG500-COOH (Rw = 0.581) in the neutral medium Fickian diffusion-controlled swelling in acidic medium and alkaline medium Anomalous diffusion and PEB-ATPEG500- COOH in acidic and alkaline media, there is a strong electrostatic repulsion, leading to slow down the relaxation rate of the polymer chain. relevant. (6) The ionic strength sensitivity test, PEB-ATPEG500-COOH ionic strength sensitivity and good salt tolerance. (7) The initial degradation experiments show that the rate of weight loss in the PEB-ATPEG500-COOH in pH7.4 PBS buffer system with time increased degradation of 98 days after the weight loss rate of less than 30%, the degradation rate is slower.
