Abstract


Artificial preparations worldwide blood supply, blood substitutes can replace human blood to perform its functions, and hemoglobin (Hemoglobin) is a major protein of red blood cells to reversibly bind oxygen carrying and delivery of oxygen to the body tissues function. Currently, blood substitutes based on hemoglobin and its derivatives, and therefore, to carry out Theoretical Study was concentrated and purified hemoglobin solution (Hemoglobin Solution) has a very important significance as a substrate for the hemoglobin blood substitutes study. Ultrafiltration as a kind of membrane separation technology, filtration pressure of about 0.01 ~ 0.5MP, use of space and has a small, low energy consumption, the advantages and the operating pressure of the pump and the pipes do not ask a small, simple device characteristics. In recent years, with ultrafiltration technology in the country's widespread popularity, ultrafiltration has been widely used in the field of bio-medicine and bio-chemical, especially used in the separation and purification of the protein. In this study, using purified hemoglobin solution ultrafiltration, to study the mass transfer characteristics of the hemoglobin solution in the ultrafiltration process, the analysis of the impact of the operating pressure on the permeate flux, mass transfer mechanism of hemoglobin solution is proposed for the process of this technology production to provide some basis for guiding. About this study: Objective: To establish the mass transfer model to optimize hemoglobin ultrafiltration operating conditions to isolate the high purity of the hemoglobin, and provide a theoretical reference for film preparation of hemoglobin. Methods: ultrafiltration concentrated hemoglobin solution the ultrafiltration concentration effects analysis under different pressure conditions, the relationship between the pressure and penetration of flux, mass transfer mechanism of hemoglobin solution in the ultrafiltration process. Results: ① under the pressure of 0.04 MPa, the temperature is 12 ℃ conditions, solution concentration were 1.69,2.07,2.66,3.66 g / L, the solution ultrafiltration permeate flux were 1.9275,1.8750,1.8125,1.7500 L/m2 · h. (2) in the same unit of time under conditions, the pressure of 0.02,0.03,0.04,0.05,0.06 MPa, ultrafiltration permeate flux solution 0.165,0.245,0.325,0.400,0.475 L/m2 · h. Better ③ test concentrated, the ultrafiltration purified hemoglobin solution than ultrafiltration former concentration increased more than 10 times, with the result of fitting experimental data want nearly was determined. Conclusion: In the process of ultrafiltration hemoglobin, membrane fluxes rendering phase characteristic relatively large membrane flux of the initial stage, after a certain time until stabilization of the membrane flux. Calculated regression of the data obtained at the same time to obtain an operating pressure in the membrane flux relationship, analog to obtain the test membrane resistance RM is 0.1182, and the pressure coefficient a is equal to 0.128. As the experiment progresses, the protein concentration gradually increased, the gel layer is gradually formed, when it reaches the gel concentration and then concentrate on the more difficult, therefore, according to the test data under the conditions of this experiment, the hemoglobin solution gel concentration Cg 18.82g / L, ultrafiltration mass transfer coefficient k is 0.7008.
