Impacts of Intense Pulsed Light on Transforming Growth Factor-beta1 Secretion in Cultured Fibroblasts and the Signaling Mechanism in the Process

Abstract


The purpose of transforming growth factor -β1 (transforming growth factor-β1, TGF-β1) signaling pathway in fibroblasts by intense pulsed light (Intense Pulsed Light, IPL) , to further explore the molecular IPL treatment of skin photoaging biological mechanisms . Isolated and cultured skin primary fibroblasts . Experiment was divided into two groups, one for the IPL treatment group , respectively, with an energy density 0 J/cm2 ( negative control ) , 10 J/cm2 , 18 J/cm2 , 27 J/cm2 , 36 J/cm2 and 36 J / cm2x2 ( 36 J/cm2 of IPL repeat treatment twice) IPL irradiation , another group for the IPL inhibitor group , including IPL ( control ) , IPL , SP600125, ( JNK inhibitor ) , SB203580 ( P38 inhibitor ) , IPL , Join inhibitor treatment after 2h , with energy density of 36 J/cm2 of IPL irradiation . Detected by ELISA in two groups of cell culture supernatant (cultural supernatants, CS) concentration of TGF-β1 , the Western blotting IPL treatment group p-JNK ( phosphorylated JNK) and p- P38 ( phosphorylation P38 ) expression. The results of the two groups CS concentration of TGF-β1 ( 1 ) IPL treatment group CS concentration of TGF-β1 : IPL treatment group CS concentration of TGF-β1 in 10 J/cm2 , 18 J/cm2 , 27 J/cm2 , 36 J/cm2 compared with the negative control decreased (P lt; 0.05), there was a significant difference compared with the negative control at 36 J/cm2 were decreased ( P gt ; 0.05 ), but no significant difference in 36 J/cm2x2 elevated compared to the negative control (P lt; 0.05), there is a statistically significant difference . ② IPL inhibitor group CS concentration of TGF-β1 : IPL SP600125, TGF-β1 concentration and control decreased ( P lt; 0.05 ) ; compared with the control , the IPL SB203580 significantly higher ( P lt ; 0.05) There were statistically significant differences . Weakened expression of p-JNK and p-P38 2.IPL treatment group : p-JNK and p-P38 signals are enhanced , 10J/cm2 strongest , 18J/cm2 began to weaken significantly weakened later . The conclusion 1.IPL inhibit the skin into the fiber cell secretion of TGF-β1 , higher energy density can promote the secretion of TGF-β1 in the lower energy density . 2.P38, JNK kinase in IPL affect fibroblasts play an important role in the process of secretion of TGF-β1 , the IPL may be raised through the JNK pathway skin into fiber cells secretion of TGF-β1 , through the P38 pathway down the secretion of TGF-β1 .
