Abstract


Objective: 1 , JS-K resistant to cisplatin-resistant human lung adenocarcinoma cell A549/DDP reversal effect . 2 , JS-K on human lung adenocarcinoma cell line A549 and cisplatin-resistant human lung adenocarcinoma cells A549/DDP multidrug resistance gene GST-πmRNA expression . Methods : MTT assay was used to detect JS-K combined with cisplatin on cisplatin-resistant human lung adenocarcinoma cells A549/DDP inhibition and reversal of multiples . 2 , using real-time RT-PCI detected before and after the treatment of different concentrations of JS-K human lung adenocarcinoma cell line A549 and cisplatin-resistant human lung adenocarcinoma cells A549/DDP in of GST-πmRNA the expression . Results: 1 IC50 values ??of cisplatin on human lung adenocarcinoma cell line A549 and cisplatin-resistant human lung adenocarcinoma cells A549/DDP (48h) is 1.702 ± 0.100μg/ml 9.943 ± 0.063μg/ml its inhibition The difference was statistically significant ( P lt; 0.05 ) ; JS- K was inhibited in human lung adenocarcinoma cell line A549 and cisplatin-resistant human lung adenocarcinoma cells A549/DDP of inhibition was no significant difference (P gt ; 0.05). JS-K for cisplatin-resistant human lung adenocarcinoma cells A549/DDP IC10 value of 1.042 ± 0.124μM . JS-K IC10 value of concentration as a non-toxic dose of 0.1μM , 0.3μM , 1μM three different concentrations of JS-K combined with cisplatin can increase the cisplatin-resistant human lung adenocarcinoma cells A549/DDP of cisplatin sensitivity to cisplatin IC50 values ??were reduced to 1.963 ± 0.238μg/ml.1.371 ± 0.119μg/ml, 1.083 ± 0.055μg/ml, the difference was statistically significant ( P lt; 0.05 ) . To cisplatin resistance reversal multiples were 5.066,7.254,9.178 2.0.1μgM 0.3μM , 1μM three different concentrations of JS-K to detect the expression of the resistance gene GST-πmRNA deal with the two types of cells after 48h , the GST -πmRNA the relative expression level decreased compared with the control group , and a dose-dependent manner , and the difference was statistically significant ( P lt; 0.05 ) . Conclusion : JS-K can enhance the sensitivity of cisplatin-resistant human lung adenocarcinoma cells A549/DDP the cisplatin reverse its resistance . This may cut multidrug resistance gene GST-πmRNA expression .
