Microorganisms under extreme environmental conditions of high salt, high temperature, high pressure, high radiation, has a unique physiological and metabolic type, the type of system development as well as unique and rich genetic resources. Total microbial DNA extracted from the the Zabuye Salt Lake sediment samples use the bacterial primers f530/r1492 amplified 16S rRNA gene, and then build the 16S rRNA gene plasmid library. Endonuclease Hae Ⅲ and Hha I positive clones plasmid DNA typing ARDRA analysis, selected clones were sequenced according to the typing results 16SrRNA partial sequence and phylogenetic tree was constructed. In the phylogenetic tree, part of the clone (57.14%) of the total number of clones with known bacteria is attributed to the same branch, are mainly distributed in the gamma-proteobacteria Gang, alpha-proteobacteria, 6 - Proteobacteria, Bacteroides door , halophilic bacterial genera, the Firmicutes and warts micro-bacteria doors, 23. The rest of the clones were uncultured sequence, a great difference with the former, formation of an independent branch of the evolutionary tree. The use of separation, enrichment culture obtained 24 isolates. Found by Gram stain, 6 bacteria are Gram-positive bacilli and cocci. 12 bacterial electron microscopic studies revealed that the isolated bacteria are rod-shaped, spherical and rod-like three forms. 14 bacterial 16S rRNA gene analysis showed that they belong to the the Firmicutes, beta-deformation Hymenomycetes and the gamma-deformation Hymenomycetes.
