Abstract


The purpose antineoplastic 5-FURP for the preparation of a model drug IL - 2 targeting 5-FURP liposomes and technology research . Liposomes (IL-2-5-FURP-L) of the IL-2 targeting 5-FURP role in IL-2 receptor expression in tumor cells , the targeting experiments , and the role of their inhibition of cell growth initial cytologic evaluation and comparison . The method using the reverse phase evaporation Preparation of IL- 2-5 - FURP -L . Liposomes 4 ℃ and 25 ℃ , respectively confined for a month , drug encapsulation efficiency , and IL-2 content as an indicator to investigate its stability . The high expression of the IL-2 receptor , cutaneous T cell lymphoma Hut-102 cells as a model cell , the MTT experiment investigated the liposome targeting and inhibition of tumor cell growth . The results by orthogonal design preparation of IL- 2 - FURP -L best prescription : drug - lipid ratio of 1:20 , phospholipid - cholesterol than 3:1 , ultrasonic time of 8 min , temperature control at 45 ° C . In the best prescription under the conditions of average encapsulation efficiency was 91.30% . Transmission electron microscope observation distribution of particle size of the liposomes uniform, the average particle diameter of about 180 nm . MTT assay results : IL-2-5-FURP-L in a dose -dependent inhibition of tumor cell growth . Free 5 - FURP ,5- FURP -L and IL - 2- 5 - FURP -L Hut-102 cells , inhibition rates were 59.19 % , 64.45% and 97.34% . UV method for the determination of IL-2-5-FURP-L of IL-2 content and drug encapsulation efficiency Conclusions In this paper , a method is accurate , simple and rapid . The stability test results show that liposomes are relatively stable when stored at 4 ℃ . Liposomes in vitro release experimental drugs through encapsulation after a good slow-release effect . MTT assay results show that IL -2 - 5 - FURP - L with the expression of the IL-2 receptor Hut -102 cells specifically bind to and play an inhibitory role better than free the 5-FURP and 5 - FURP -L . Whereby instructions the ,5-FURP prepared into IL-2 targeting 5- FURP liposomes , of IL- 2 play in the TDDS in the role of the guide is , to achieve the targeted release of drugs at the cellular level , so IL-2-5- FURP-L to inhibit tumor cell growth and enhanced role .
