Abstract

The first part observed using polarized light microscopy before and after fertilization and abnormal fertilized egg zona pellucida research purposes using polarized light microscopy (polarized light microscope, Polscope) observation and measurement of human oocytes and zygotes transparent with density (the zona pellucida density ZPD), zona thickness (zona pellucida thickness, ZPT), analysis of changes in vitro maturation process of the change of the zona pellucida (zona pellucida, ZP) and after fertilization ZP, the different insemination after fertilization zygote ZP difference and the same in vitro fertilization method (in vitro fertilization , IVF) ZP different fertilization outcome correlation. 18-20 h after IVF fertilization of methods using polarized light microscopy 18-20 h after the 68 cases in 248 fertilized eggs and 60 the immature eggs camera; intracytoplasmic sperm microinjection (Intracytoplasmic sperm injection, ICSI) fertilization 61 of the 11 cases, a fertilized human egg camera; 36 ICSI cycles 127 eggs, egg remove 2-4 h after removal of granulosa cells, camera. ZP, software measurement of the captured images to analyze the changes in the maturation process and after fertilization of ZP, the difference of the differences of the different methods of fertilization ZP and different fertilization outcome of ZP. Results the immature oocytes layer ZPT inner layer ZPD and ZPT higher than the mature egg after fertilization, IVF layers ZPT, ZPD was significantly increased (2.49 ± 0.49, 8.79 ± 1.91, 19.33 ± 2.28 vs. 2.14 ± 0.52, 7.98 ± 1.73 , 16.54 ± 2.43; p lt; 0.05); IVF and ICSI fertilization, the fertilized egg ZP significant difference between prokaryotic (3 pronuclear, 3PN); IVF fertilized egg inner ZPT lower than 2PN (8.19 ± 1.68 vs. 8.79 ± 1.91 ; p lt; 0.05), 0PN whole layer and the inner layer ZPT, ZPD higher than 2PN (2.81 ± 0.69 and 9.55 ± 1.45, 20.79 ± 3.41 vs. 2.14 ± 0.52, 7.98 ± 1.73, 16.54 ± 2.43; p lt; 0.05 ). Egg density and thickness of different conclusions maturity, the ZPT, ZPD changes after the egg is fertilized; the different insemination ZP differences; fertilized egg ZP structure correlation with fertilization outcome. The second part 6h short time insemination with 18h overnight inseminated zona pellucida change random self-control research purposes fertilized egg ZP and embryo quality between 6 h and 18 h of sperm-egg coculture group relations approach 21 cycle 258 eggs randomly assigned to 6 h 18 h sperm-egg co-cultured group of self-control, and 20 h after fertilization polarized light microscope camera, analysis of normal fertilized egg ZP and embryo quality differences between the two groups. Results 6 h and 18 h groups, fertilization rate and polyspermy rate differences portable embryo rate is slightly higher (70.1% vs 63.27%) no statistical differences the transferred embryos proportion (67.80 vs. 37.21%) . 6 h group of normal fertilized egg inner layer, full-thickness zona pellucida thickness (zona pellucida thickness, ZPT), inner transparent with density (zona pellucida density, ZPD) (8.28 ± 1.28, 18.97 ± 2.64, 2.09 ± 0.50vs. 8.86 ± 1.52, 19.57 ± 2.66, 2.24 ± 0.67) less than 18 h group; development of portable embryos fertilized egg ZPD (2.07 ± 0.49 vs. 2.26 ± 0.72) was significantly lower than the 18 h group, the non-transferred embryos develop into The fertilized egg outer layer ZPT (9.14 ± 2.46 vs. 7.74 ± 1.37) higher than the 18 h group. Fertilization combination of sub-transparent conclusion is short with inner layer thickness, density than the the traditional overnight fertilized combinator low, which may be beneficial embryos hatched.
