Abstract

Every year soybean straw is rich in our country, but its feed efficiency is lower and the resource waste is very severe. The high crude fiber content and lignification degree, low crude protein content are the main limiting factors to widely using for animal feed of mature soybean straw. The goal of this research was to understand genetic mechanisms that control the quality of soybean straw.Toward this end,We have undertaken an analysis of the accumulation law of development, genetic mechanisms and gene mapping on protein and ligin. Some molecular markers tightly linked to the aimed target traits are obtained and thus would be helpful on quality improving of soybean straw via molecular marker-assisted seleetion (MAS).This has important theoretical and praetieal significance on enhancing international market competitiveness of soybean straw in our country. The main experimental results are as follows:First, with the major gene plus polygene model of quantitative traits,a inheritance analysis of NJRIKY was carried out to investigate the inheritance of the protein. Based on the genetic research above, QTL mapping for protein was carried out as following. The mapping Population with 184 RIL plants of the cross KEFENG×NANNONG1138-2 was established and tested for their genetic variation, and then used to construct a genetic linkage map spanning 25 linkage groups. The result showed that the inhetitance of the protein fitted two major genes plus polygene modle. Hetitability values of the major genes of the RIL were estimated as 82.05%,78.36%respectively,while the heritability values of those polygene were lower. In the improvement of straw quality, accumulating polygenes should be emphasized, while utilization of majo rgenes is preferred. The methods of composite interval mapping (CIM) of software Win QTL Cartographer Version2.5 were used for QTL Mapping. With CIM method, eight QTLs were mapped on 4 linkage goups:A2, Dla, C2,G,explaining 7.89%—17.02% of the total phenotypic variation.Second, with the major gene plus polygene model of quantitative traits,a inheritance analysis of NJRIKY was carried out to investigate the inheritance of the ligin. Based on the genetic research above,QTL mapping for ligin was carried out as following. The mapping population with 184 RIL plants of the cross KEFENG×NANNONGl 138-2 was established and tested for their genetic variation, and then used to construct a genetic linkage map spanning 25 linkage groups. The result showed that the inhetitance of the ligin fitted two major genes plus polygene modle. Hetitability values of the major genes of the RIL were estimated as 74.14%,92.97%respectively, while the heritability values of those polygene were lower. In the improvement of straw quality, accumulating polygenes should be emphasized, while utilization of majo rgenes is preferred. The methods of composite interval mapping (CIM) of software Win QTL Cartographer Version2.5 were used for QTL Mapping. With CIM method, eleven QTLs were mapped on 4 linkage goups:A2, B1, C2,M, exPlaining 3.11%-10.12% of the total phenotypic variation. The results of inheritance mechanism of quantity of straw, obtained by major gene and polygene mixed inheritance model and by QTL mapping, are relatively in consistency. For the number of quality gene and their exact positions still was unknown. The results of this research are preliminary and tentative.and should be studied in the successive research work.
