Abstract


Tubulin is generally expressed in a eukaryotic cell protein, tubulin (TUA) by α and β-tubulin (TUB) consisting of tubulin heterodimers constituting . A total of six different TUA genetic subtypes and 9 different TUB gene isoforms in Arabidopsis . Many studies have confirmed that different tubulin gene isoforms highly expressed in a specific developmental stage or tissue , will also change with the external environment stress expression levels . Therefore , microtubule protein gene just housekeeping genes , and is also involved in many signal responses . The the study TUA2 genetic subtypes expression changes mutants for material . Use of reverse genetics method , the use of genetic transformation Tail PCR , semi- quantitative PCR technology TUA2 involved in ABA stress on seed germination process . The main results are as follows : 1 . Physiological detection in the number of T-DNA insertion mutants discovered a mutant sensitive to ABA . The method find use Tail PCR upstream promoter region of the T-DNA insertion site is located TUA2 gene and found that the locus of insertion result in mutant TUA2 improve gene expression . In addition, we find the appropriate sites mutant SALK library . Three different strains through genetic transformation method TUA gene overexpression body . 2. Constructed the subcellular localization TUA2 gene transgenic expression vector , and the use of a gene gun into the TUA2 gene and GFP fusion gene expression in onion epidermal instantaneous , The results show TUA2 mainly in the plasma membrane distribution . 3 by the detection of the the ABA stress under seed germination rate found TUA2 gene mutations body , super express body delay seed germination . The speculated TUA2 may be involved in the regulation of ABA on seed germination process . 4. ABA pathway gene expression by the RT-PCR method , a result of analysis , compared with wild-type mutants of HAB and ABI gene expression was significantly reduced in the ABA treatment , suggesting TUA2 may be involved in the ABA negative regulation of signal system process .
