Abstract


In this thesis, Lop Nur region archaea for the study, to be solved for Lop Nur region archaea can cultivate a diversity of scientific problems. Learn from the experience of separation halophilic actinomycetes, screened for the separation medium, pure culture techniques as a means, combined with a system based on the 16S rRNA gene phylogenetic analysis, research Lop Nur region halophilic Archaeon species diversity; through three kinds of functional enzyme screening, preliminary to explore the the halophilic archaea potential value. In order to further study the Lop Nur region archaea resources and development by providing technical support and theoretical basis. Lop Nur region of 13 soil samples collected from the selection of the CCMS improved Gause ISP 5 HV isolation medium isolates purified to repeat a total of 56 archaea. Phylogenetic analysis of 16S rRNA gene system, found that 56 Zhu Zhu archaea new genus distributed in five candidate: Group 1, Group 2, Group 3, Group 4, Group 5 and 10 take effect publication is: salt marine bacteria genera (Halopelagius), sodium lines genus (Natrinema), salt the the terrestrial genus (Haloterrigena), salinity group coccus (Halalkalicoccus), salt idler genus (Halopiger), appropriate salt genus (Halodaptatus), salt red bacteria genus (Halorubrum), salinity coccus (Natronococcus), salt stack coccus (Halosarcina),, Salt Lake habitat genus (Halostagnicola). Initially identified five candidate new genera and 38 new species candidate, Lop Nur in Xinjiang region contains a wealth of of halophilic archaea new species resources; Shannon index of 1.795, indicating that in the region there is a huge archaea mining resources. The different results show that the rate of medium out bacteria: the CCMS medium than improved Gause 1 medium; different medium separation of species diversity results show that: CCMS better than Gause 1 medium. Description CCMS medium the halophilic archaea separation is better, for further study of the Lop Nur region archaea resources to provide technical support and guidance of the idea. Selected according to the different branches of the phylogenetic NJ tree the 17 archaea salt concentration in the experimental and functional enzyme screening. The salt concentration of the experimental results showed that the growth of most strains of the range of 10% -35%, the optimal salt concentration of 20%; amylase, protease, esterase three kinds of functional enzyme activity rate detection result: amylase activity detection rate of the strains 70.6%, the protease activity strain detection rate of 35.3%, esterase activity strain detection rate of 82.4%. The halophilic archaea enzyme activity higher detection rate and strong activity in some strains, and provides a theoretical basis for the further development and utilization of archaea resources. Polyphasic approach, YIM 93701 and YIM 93664 from the morphological, physiological and biochemical characteristics of the chemical composition of the tripartite face identification. The results of preliminary judging YIM 93701 and YIM 93664 salinization corporation the coccus (Halalkalicoccus) candidate new species.
