Biosurfactants (Biosurfactants), refers to the microbial culture under certain conditions resulting surface active substance is a natural surfactant. It addition to the general surfactant to reduce surface tension, increase the stability of emulsions and foams and other effects, both non-toxic, biodegradable characteristics. Because biosurfactants these features, so it has a wide range of applications. Therefore, for screening culture substrate inexpensive and stable performance of the high-yield strains, cost-effective product separation extraction method is biosurfactants an important research direction. In this paper, oil spreading method and the method of screening blood agar, isolated from oil sludge screening can get 11 strains produce biological surfactants. Through its surface tension of the liquid broth further validation, pick out a good performance and stability and the surface Zhang Liliang strains which numbered N-①. Then, the extracted DNA genome sequencing 16SrDNA through the strain, according to the NCBI Genebank database homology with the strains of recent taxonomic status of strains and isolates of sequence information for phylogenetic analysis, software generation system through MEGA3.1 phylogenetic tree, combined with bacterial physiological and biochemical tests, the strain was identified as Bacillus subtilis (Bacillus subtilis). To ensure follow-up experiments and strains during fermentation remains optimal culture conditions, the drain ring size as the main indicator of their original culture medium was modified. Improved medium was (g / L): NaCl 20g; KH2PO4 5g; MgSO4 · 7H2O 0.5g; peptone 1g; Glucose 20g; pH 6.0, medium volume 80mL/250mL; inoculum size of 5%. Then the strain N-① biosurfactant produced materials were extracted, the extracted material obtained by the color reaction and infrared spectroscopy to determine the surface active agent is a peptide lipid surfactant. And the strain N-① the biological properties of surfactants were determined. The results show that the surfactant which produces the critical micelle concentration of 130mg / L; has good emulsifying properties which can maintain the volume of diesel and water-emulsion at about 64%; with strong scope of its temperature, pH, NaCl tolerance ranges of 20 ℃ - 80 ℃ ,4-11, 1% - 8%. Biosurfactant producing bacteria with its wide range of applications, the paper conducted a preliminary study of its application by the wax experiments we can conclude that after surfactant treatment paraffin its structure from the original compact, becomes a sparse structure. While producing bacteria treated with a surfactant petroleum-contaminated soil, the results show that the oil-contaminated soil in the oil degradation rate of 64%, indicating that the surfactant producing bacteria on petroleum contaminated soil has degrade performance.
