Melatonin (Melatonin, MEL) is an important endocrine hormone secreted by the pineal gland and retina, it is widespread multiple organizations in multiple species, an important signal lighting information in coordinating relations with the reproductive system, the light on the regulation of reproductive function rely melatonin mediated, in combination with the melatonin receptor (Melatonin receptor, MTNR) to adjust the animal's circadian rhythm and reproduction changes. The purpose of this study is to understand the the chicken of MTNR gene's genetic variation from the molecular level and open prenatal differential expression of different organizations, in order to provide the basis for the genetic improvement of local chicken breeds. The results are as follows: Wenchang Chicken as the object of study, designed pairs of primers, the use of cloning and sequencing and direct sequencing methods, were amplified 5'UTR of the three MTNR1A, MTNR1B and MTNR1C receptor gene and exon regions, with Wenchang Chicken egg production traits do correlation analysis. The results show that there are 14 SNPs and one indel sites: MTNR1B receptor gene 5'UTR. -778 Locus mutation gene (A → G) of the egg production of the the Wenchang Chicken medium-term (22-31 weeks) and 46 weeks of age the dominant effect of the total egg number were 2.3 and 6.2. Late (32-46 weeks of age) egg number and 46-week-old egg number -216 locus C of A is the dominant allele, additive effects were 2.5 and 3.2; dominant The effect of 3.5 and 6.8, respectively, and showed overdominance. The two loci haplotype analysis showed that AA single-type total egg number to 46 weeks of age effect values ??in the lowest of the four haplotypes; AA / AA merger genotype of the hen's age at first egg was significantly later in AC / AA and GC / GC genotype (P lt; 0.05). Of MTNR1A, MTNR1B receptor gene exon is highly conserved, and on the existence of in MTNR1C receptor gene exon3 in two C378T and G381A SNPs, but these two sites did not result in an amino acid change. Β-actin gene as an internal standard, using qRT-PCR method, compared of MTNR1A, MTNR1B and MTNR1C genetic differences in the heart, liver, fallopian tubes, ovaries, brain and optic chiasm organization and expression of the chicken open prenatal. The results show that: of MTNR1A gene in different organizations open postpartum showed upregulation (except brain expression down outside), no significant differences in expression levels in the open pre-natal and post-opening prenatal organization, but the organizations in the opening postpartum in the expression of a significant difference, and the expression level of the highest to the ovary. Compared with the open prenatal the organization open postpartum MTNR1B, and MTNR1C gene in addition showed upregulated in the optic chiasm, the expression levels of the rest of the organization presented a downward trend. Amount the MTNR1B gene expression in the same organization open to prenatal differences were not significant, but the expression of different organizations open prenatal or postnatal open significant difference and are suprachiasmatic highest expression level was the lowest in the ovary; and MTNR1C genes in addition to expression in the different tissues open prenatal or postnatal open significant differences, there are significant differences in the heart, the fallopian tubes, brain tissue after open prenatal. The results of this study for further research on the the chicken breeding performance melatonin receptor gene regulation mechanism to provide reference.
