Abstract


ObjectiveExperimental study of Neuropeptide Y and in the pathogenesy of cervical vertigo; The relationship between analysis of cervical instability and neuropeptide Y; To provide a indicator is to early of discovery and treatment,and improve the diagnosis of cervical vertigo.MethodsChoose 30 healthy adult Japanese white rabbits as the research object, and these rabbits were randomly divided into three groups (n=10): the experimental group were established animal model of cervical vertigo caused by cervical instability and chronic vertebral artery insufficiency through anterior and posterior′s surgery; sham operation group only incision surgery; control group without any surgery. Experimental groups were respectively collected venous blood 2ml before surgery, 3 days, 30 days, 60 days, 90 days after surgery to learn rabbit neuropeptide Y (NPY) levels by ELISA(enzyme-linked immunosorbent assay) measurements. Appliy improved Rivlin law (slope test) to detect changes on the location and their own sense of space and balance; after 90 days experimental groups were respectively used cervical spine X-ray examination to observe the relative mobility of the fourth vertebra, hemodynamic check to observe cervical vertebral artery pulsatility index (Pi), resistance index (Ri) and vertebral artery blood flow changes; Production of the vertebral artery biopsy after rabbits were killed to observe of cervical artery diameter (ED), inner diameter (ID), wall thickness (WT) changes. The data collected using statistical software SPSS16.0 Processing.ResultsUsing the one-way ANOVA of 90 days after surgery, the relative mobility of the fourth cervical vertebrae ,the vertebral artery pulsatility index, resistance index, wall thickness of surgical group were higher compared with the sham group and control group, the angle based on improved Rivlin method (slope test), the vertebral artery blood flow and vertebral artery diameter of surgical group were less than the sham group and control group, and sham operation group and control group were significantly difference (P <0.01 ), while vertebral artery diameter between the sham group and control group were not significant difference (P> 0.05), and so was sham group and control group before and after surgery (P> 0.05) ; using the repeated measures analysis of variance of NPY level determination: 3 days after surgery, NPY content of surgical group and sham group was significantly increased compared with before surgery, it was significantly statistical difference (P <0.01), while no significant difference in the control group (P> 0.05); 30 days after surgery , NPY levels of surgical group were significantly reduced (P<0.01) compared with the sham group after 3 days.And difference was more obvious between the sham group and the surgical group (P<0.01), and both were higher than the control group (P<0.01); 60 days after surgery, NPY level of surgical group had started to rise slowly compared with 30 days after surgery (P<0.01), while NPY level of the sham group and control group were unchanged (P> 0.05); 90 days after surgery, NPY level of surgical group had continued to gradually increase compared with after 60 days (P<0.01), while NPY levels of the sham group and control group were unchanged (P>0.05), difference between any two groups were statistically significant (P<0.01); using the linear regression analysis of factors related to cervical vertigo: the angle based on improved Rivlin method (slope test) as the dependent variablity was significantly affected by NPY level and diameter of the vertebral artery as independent variablity, P values were 0.028, of which the slope angle was positively correlated with vertebral artery diameter.Conclusion1.Through the Injury by anterior surgical injury C4/5 anterior longitudinal ligament and intervertebral disc structure, surgical ligation and cut off the Muscles of C4/5 segment supraspinous ligaments, the method successfully simulated cervical spine instability and such as long bow and degeneration risk factors, the ability to produce of animal models of cervical vertigo.2.The increase of neuropeptide Y is more prone to vertebral artery stenosis. not only direct and indirect enhancement of norepinephrine induced contraction, and to prolonged the Contraction time,in late vascular smooth muscle cells (VSMC) proliferation,the neuropeptide Y is the main factors of the Vertebral artery insufficiency.
