Abstract

Rat restraint - soaking stress (the restmint water-immersion stress RWIS) process that causes stomach ulcers, supraoptic nucleus, paraventricular nucleus of the medulla oblongata of fans take the complex nuclei of neurons involved in the regulation. These neurons involved in the reaction, arginine vasopressin (AVP) can, oxytocin (OT), catecholamines (NE) neurons have been identified, but they account for only a part of the neurons involved in the reaction. The remaining neurons is what type? Opioid peptide neurons is involved in the reaction? Have not been reported. Previous studies have shown that many parts of enkephalin in the striatum, hypothalamus, diencephalon, brain stem and spinal cord are distributed in the hypothalamus, enkephalin usually coexist with AVP, OT neurotransmitter. Intraperitoneal injection methylthio - enkephalins analogues, cold restraint stress induced gastric mucosal injury prevention. Methylthio - enkephalin (M-ENK, opioid peptides a) have an important role in the regulation of gastric mucosal injury. Therefore, this study uses a double standard and intracerebral microinjection technology Met - enkephalin Fos protein immune explore whether methylthio neurons involved in RWIS reaction - enkephalinergic neurons, the neuronal activity corresponding stimulated gastric mucosal injury due role. The study is divided into four parts: the first part, different methods of pretreatment expression of M-ENK RWIS rat central. Respectively, by intraperitoneal injection of saline, youthful Sin prime 1mg / k (g body weight) and intracerebroventricular injection of colchicine prime 100μg/kg (weight) of the method pretreatment rats, observed the different processing methods for rat M-ENK Immunohistochemical positive expression. Found that intraperitoneal injection of intracerebroventricular injection similar effect could obviously shows the cell bodies of neurons. The saline group can not be showing the cell body. Thus, we chose the intraperitoneal injection method is simple follow-up test. The second part, RWIS rat hypothalamic expression of M-ENK in the medulla oblongata. Single labeled by immunohistochemistry and double-labeling technique to determine RWIS supraoptic nucleus, paraventricular nucleus, brainstem vagal composite body M-ENK positive expression of M-ENK Fos protein expression in the case. The results show, the vagus Dorsal nuclear, nucleus of solitary tract suspected nuclear have significant M-ENK immune positive expression, but the stress group and control group comparison, no significant forward difference (P gt; 0.05); regardless of M-ENK immune response single standard, or M-ENK Fos immune double standard, supraoptic nucleus, paraventricular nucleus no positive cell body. Tip in the RWIS 1, the expression of the medulla oblongata M-ENK no significant influence, no significant effect on the activities of the M-ENK neurons in the medulla oblongata role. In addition, the distribution of M-ENK nerve endings in the striatum, experimental and control groups showed no significant difference (P gt; 0.05). The third part of the lateral ventricle microinjection of M-ENK RWIS induced gastric mucosal injury in rats. To using the lateral ventricle pipe, microinjection of M-ENK (10ug/5ul / only) the naloxone (10ug/5ul / only) and then carried RWIS observed gastric ulcer caused by the M-ENK and naloxone RWIS produce significant impact. The results showed that gastric mucosal damage index between the M-ENK group, the Naloxone group and the control (saline) group showed no significant differences (P gt; 0.05), prompted by exogenous and endogenous M-ENK gastric mucosal injury caused on RWIS no significant impact. The fourth part, RWIS Fos protein expression in rats several endocrine glands. By immunohistochemical methods, detection of the pineal gland, thyroid, adrenal, pancreas bondage - dip stress in Fos protein expression situation. Results show that, compared with the control (no stress), the experimental group in the thyroid in the adrenal medulla and the cortex zona Fos protein-positive cells number and accumulated optical density (IOD) increased significantly (P lt; 0.05) in the experimental group and the control group; pineal Fos protein expression in the experimental and control groups are strong, obvious differences between the two groups (P gt; 0.05); adrenal cortex zona reticularis islets were not found Fos protein expression. Prompted rat thyroid, adrenal zona glomerulosa and medullary cells are involved in the the RWIS response. In summary, M-ENK positive neurons in the hypothalamus and medulla oblongata observation cell body is only found in the nucleus of solitary tract; dorsal vagal nucleus of the medulla oblongata, nucleus ambiguus immunoreactive nerve fibers in the peripheral distribution; final area, paraventricular nucleus, supraoptic nucleus of M-ENK-positive neuronal cell bodies and fiber endings; M-ENK distribution of nerve endings in the striatum. RWIS of the medulla oblongata solitary tract nucleus neuronal activity of M-ENK not significant; dorsal motor nucleus of the vagus nerve suspected nuclear M-ENK-positive nerve endings activities impact is not significant. M-ENK content changes within the central Gastric Mucosal Injury bondage flooding major endocrine gland activity not play a major role.
