Objective: dextran sulfate sodium (DSS)-induced experimental ulcerative colitis (UC) by observing Clostridium mouse colon mucosa of intestinal trefoil factor (ITF), nuclear factor-kappa B (NF-κB) and tumor necrosis factor-α (TNF-α) expression of Clostridium role of UC and possible mechanisms. Methods: 60 male BALB / c mice, 6-8 weeks of age, body weight 19.0 ± 2.0g, clean grade. Randomly divided into six groups, were the normal group, model group, negative control group, positive control group, the Clostridium low dose group, Clostridium high-dose group (n = 10). Normal group free drink distilled water, and the rest of the group drinking 3% DSS (molecular weight 36,000 -5 million) seven days to establish a mouse model of acute UC. Modeling began the next day, in addition to the normal group, the remaining groups simultaneously given an empty gavage needle sterile saline mesalazine 29.80mg/ml, and Clostridium broth 1 × 106CFU/ml 1 × 10 7 CFU / ml gavage, 0.2 ml / 2 times / day. During the experiment to observe and record the general condition of the mice, weight, stool occult blood experiment eighth day with fold neck Act, the mice were sacrificed, free and remove the whole colon, measuring back the ileocecal to anus length. Portion of colon tissue and then wash with cold saline colon fixed in 4% formaldehyde, embedded in paraffin, HE staining; remaining colon tissue preservation at -70 ℃, soluble in production organization. The observed indicators include: disease activity index (DAI), colon length, histological injury score; the immunohistochemistry assay colon tissue ITF and NF-κB expression; radioimmunoassay detect the expression of TNF-α. Results: 1. DSS drinking after each intervention the mice are varying degrees of diarrhea, mucous, bloody stools and other symptoms, with the increase in the number of days in mice DAI integral gradually increased. Clostridium can improve the general condition of the mice, to reduce mice DAI integral histologic injury score, a statistically significant difference compared with the model group (P <0.05). 2. Compared with the model group and negative control group, Clostridium mesalazine treatment group UC ITF expression in the mouse colon mucosa increased (P <0.05); decreased expression of NF-κB and TNF-α (P <0.05) . Clostridium results have been very low-dose group and positive control group (P> 0.05). Conclusion: DSS-induced acute colitis can simulate the clinical characteristics of the attack of UC. Clostridium treatment of acute UC, efficacy and dose related, part of its mechanism may be related to the Clostridium can increase the expression of the ITF, the inhibition of NF-κB and TNF-α expression.
