Objective: To compare recombinant human vascular endothelial inhibin, the PYM used alone and combined effects of human umbilical vein endothelial cell line HUVEC cells to explore the related mechanism of action, to reveal endostatin is applied to the treatment of proliferating hemangiomas feasible sex. Method: pass cultured HUVEC cells, take 3-5 generation cells grow well, different concentration of the drug endostatin, BLM acting on HUVEC cells, MTT (MTT), comparing the two drugs for 24h changes in cell morphology before and after the drug action, the immunohistochemical assay before and after the drug effect HUVEC cells within the vascular endothelial growth factor (VEGF cell inhibition rate of HUVEC cells, 48h, hematoxylin - eosin (HE) staining light microscopy ) positive differences in the expression, flow cytometry analysis of drug action HUVEC cell apoptosis. Results: Endostatin Pingyangmycin respectively role HUVEC cells after the absorbance value (OD) with the concentration of the drug decreases, the two drugs on the inhibition of HUVEC cells showed a concentration-dependent and time-dependent . VEGF immunohistochemical staining showed that the two drugs were role HUVEC cells cytoplasmic gray value increase. Light microscopy showed: HUVEC cell morphology the endostatin role after significant changes, coverslips can see a large number of apoptotic cells, and increased with the increase in drug concentration, PYM after a large number of cell suspension, the number of adherent cells reduced, the greater the concentration of the drug and less adherent cells. Flow cytometry two drugs used alone will cause HUVEC apoptosis, and showed more for the concentration, the greater the number of apoptotic cells; two drugs used in combination showed synergistic inhibitory effect on HUVEC cells. Conclusion: Endostatin, BLM HUVEC cells were significantly inhibited, can cause apoptosis, and preventing VEGF into HUVEC cells, thereby inhibiting cell proliferation, the two drugs on the inhibition of the vascular endothelial cells related mechanism there is some commonality; higher concentrations of endostatin apoptosis is still the main PYM concentration is greater than a certain range (gt; 20μg/ml) cells inhibit gradually necrosis dominant; Pingyangmycin vascular endothelial the cells adherent than endostatin obviously reveal the difference of the two drugs on the inhibition of the vascular endothelial cells exist; the endostatin pingyangmycin joint use a synergistic effect on the inhibition of endothelial cells, for two drugs used in combination treatment of proliferating hemangiomas provide the basis of the basic theory.
