Abstract


Objective: To determine a hereditary hemochromatosis pedigree (A) and its paralogs (B) clinical MRI features and liver hepcidin mRNA expression to explore the pathogenesis of the disease may be . Methods: A, B family members for clinical , biochemical , liver histopathology, multi-organ MRI examination and qRT-PCR assay hepcidin mRNA expression in liver tissue . Results : (1) A portion of the sub family members hemochromatosis clinical manifestations and biochemical abnormalities , B sub- family members were related to clinical manifestations, some members of the biochemical mildly abnormal ; ( 2 ) 3 patients underwent liver biopsy and found two cases are more iron deposition in liver cells , and the other one case showed no iron deposition ; (3) MRI: A, B pedigree liver, spleen pancreas some members have different degrees of iron deposition , showed low signal , and to T2-weighted images are more sensitive to the kidneys , the brain showed no iron deposition . (4) qRT-PCR assay underwent liver biopsy in three cases of patients diagnosed with liver hepcidin mRNA upregulation . Conclusions: ( 1 ) The Scarlet iron pedigrees were deposited mainly in the liver , other organs such as the heart , spleen , pancreas are also involved ; while its collateral visible part of the members of the biochemical abnormalities and organ iron deposition , but has not yet found the relevant Scarlet clinical manifestations of disease , whether it is in the incidence of early yet to follow-up observation . ( 2 ) Compared with liver biopsy , MRI tissue iron deposition for more accurate diagnosis . ( 3 ) liver hepcidin mRNA expression increased , suggesting that serum hepcidin levels , suggesting that there may hepcidin resistance, thus affecting negatively regulate hepcidin role , which led to the occurrence of iron deposition in the body .
