Abstract


In this study of 279 samples were collected in the suspected flu symptoms swine flu virus separation from Jilin Province in 2007-2008; using hemagglutination, hemagglutination inhibition, neuraminidase inhibition assay virus subtype identification. Three swine H3N2 subtype of influenza A virus antiserum was prepared for the analysis of genetic variation in Jilin Province swine H3N2 influenza virus and antigenic analysis. The results showed that the three swine H3N2 virus antigen is not exactly the same, these virus antigenic change is occurring or has occurred. HA amino acid sequence analysis showed that the sites of three pigs H3N2 influenza virus HA antigen, the receptor binding site, the glycosylation sites at amino acids there are different degrees of mutation. The phylogenetic tree analysis showed that the swine H3N2 viruses isolated in 1998 and 2008, and of modern human influenza virus genetic relationship, in which the M of A/swine/Jilin/37/2008 and NS gene with avian H5 virus in the same evolutionary branch. Jilin Province, there are not only pigs the Modern Human lineage viruses, and the lineage of modern man first discovered the virus and the avian H5 influenza virus gene rearrangement events exists. The use of solid-phase enzyme-linked three swine H3N2 influenza virus receptor binding characterization results show that the three virus also has the ability to bind with SAα2, 3Gal and SAα2, 6Gal two types of receptors but SAα2, 3Gal receptor binding capacity than SAα2, 6Gal receptor. Description of these viruses in maintaining the receptor binding properties of the avian influenza virus, and to obtain and express SAα2 6Gal receptor binding ability, so as to have a greater potential infection. The histopathological observations show that has the avian H5 internal genes A/swine/Jilin/37/2008 Human lineage within the A/swine/Jilin/5/2007 compared, the former weight of the BALB / c mice The more obvious changes in the severity of lung lesions stronger than the latter. Mammalian H3N2 virus subtype avian H5 virus internal genes enhanced virulence. The study suggests that there are different antigen subtype H3N2 virus, in Jilin Province herd mainly Modern Human lineages virus, because of the swine flu virus also has the ability to bind to two types of receptors within the lineage, increase the risk of infection. Modern man first discovered the source lineage the virus avian H5 subtype virus natural rearrangement fact, the virulence of the virus of BALB / c mice enhanced. Therefore, strengthen the swine flu epidemiology and molecular epidemiology survey and research, has important implications on the master influenza virus Genetic variation and prevention of animal and human influenza.
