Abstract


Objective The purpose of this study was to examine the changes in number of endothelial progenitor cells (EPCs) from umbilical vein blood in placental vascular disease(abnormal umbilical blood flow S/D ratio) patients.Methods 18 women with abnormal umbilical blood flow S/D ratio and 21 normal pregnant women in the third trimester were studied.Mononuclear cells(MNCs) from umbilical cord vein blood were isolated by Ficoll density gradient centrifugation. EPCs were cultured in vitro and re-suspended in M-199 medium supplemented with 20% fetal bovine serum and human vascular endothelial growth factor and basic fibroblast growth factor, culturel dishes were coated with fetal fibronectin before culture.3 and 7 days later, the cells were identified by positive expression of both CD133 and CD34 under laser scanning confocal microscope and determined the number of EPCs with flow-cytometry.Results The results showed that, compared with normal pregnant women, the number of EPCs was decreased relatively when just isolated from umbilical vein blood in placental vascular disease patients after being cultured for 3 days and 7 days. The number was (13.55±1.54)% vs. (5.13±1.36)% and (63.24±3.24)% vs. (36.17±1.45)% and (81.57±3.27)% vs. (59.48±2.51)%, respectively(P<0.05).Conclusion It is concluded that the number of EPCs was decreased relatively when just isolated from umbilical vein blood in placental vascular disease (abnormal umbilical blood flow S/D ratio) patients.
