Abstract


Alzheimer disease (AD) is one of the most common neurodegenerative diseases. Along with the accelerated speed of poppulation aging in China, there is a drastic increase in the incidence of AD. It makes up approximately 70% of all dementia cases. There exist two major distinctive hallmark lesions within the disessed brain, which are neurofibrillary tangles compsed of hyperphosphorylated Microtuble-associated protein tau and senile plaques coposed ofβ-amyloid. In addition, Neuronal loss appeared widely in diseased brain and atrophic with widened sulci and narrowed gyri. But the pathological mechanism of it has not been elucidated.It is widely agreed that AD is shaped by interactions between genes and environment factors. The vast majority of the cases are sporadic forms, likely resulting from complex gene-gene and gene-environment interplay. Dosunmu R thought that genetics play a minor role in the etiology of AD, so it is more important to investigate the environment and diet as primary risk factors in AD pathology. More and more studies show that several environmental factors, including pesticides, air popllution, metals and head injuries, lifestyles and dietary habits have been associated with increased disease risk or even with protection. Schulte invetigate that neurodegenerative diseases gathered in pesticides pxposure、organic solvent and electromagnetic field group-workers. And there is also a report that occupational exposure to pesticide increases the risk of incident AD. Pesticide exposure has obvious neurotoxicity and effects on early neurobehavioral development and postnatal growth of wistar rat offspring. But the exact pathological mechanism about the relationship between pesticide exposure and AD has not been elucidated.Pesticides have been widely used in agricultural production. So it is necessary to detect its danger. Here, we studied the effects of pesticides exposure on the ability of learning and memory of male SD rats and its underlying mechanisms. Compared to the cntrol group, we found that there exsits a memory deficit during the pesticides exposure groups, and hyperphosphorylated tau levels increase. We also found an imbalance between glycogen synthase kinase-3 beta and protein phosphatases (PP) 2A which play important roles in reglating the phosphorylation level of tau protein.
