Objective: To study the hepatitis B virus X protein (HBX) through AKT signaling in the regulation of the expression of c-Jun protein in hepatic oval cells . Methods: by fluorescence microscopy , immunofluorescence , Western Blot identification of HBX protein expression in pEGFP-HBX-LE6 cells . 2 the pEGFP-HBX-LE6 oval cells as the experimental group , L02 - HBX cells as a positive control group , the LE6 oval cells , oval cells transfected with empty vector pEGFP-LE6 general control group . Western Blot Detection of p -Akt , AKT and c-Jun protein expression levels . 3 PI3K/AKT pathway inhibitor LY294002 to blocking PI-3K/AKT signal path , by Western Blot detection of changes in the level of c-Jun protein AKT pathway is inhibited . Results: 1. HBX genes stably transfected with pEGFP-HBX-LE6 transfected with empty vector pEGFP-LE6 Oval Cell Line visible green fluorescence under a fluorescence microscope ; HBX protein in pEGFP-HBX-LE6 cells express ; cells immunofluorescence the HBX expression the pEGFP-HBX-LE6 oval cells , and is mainly located in the nucleus . 2.p-Akt, c-Jun protein in pEGFP-HBX-LE6 cells upregulated . 3.AKT pathway inhibition HBX upregulation of c-Jun protein in the pEGFP-HBX-LE6 subsequently lowered . Conclusion : HBX hepatic oval cells raised the AKT signaling and c-Jun protein expression , and HBX protein through upregulation of AKT signaling protein expression of c-Jun .
