Abstract


[Objective]: PA-MSHA mouse pancreatic cancer xenograft model as a research object, observation of Pseudomonas aeruginosa preparations (Pseudomonas Aeruginosa Injection PA-MSHA) inhibition of the growth of the mice pancreatic cancer xenograft model, discussed in inhibition of the mouse pancreatic transplantation tumor growth and proliferation mechanism of action, effective for the treatment of pancreatic cancer explore new methods. [Method]: in mouse pancreatic cancer cells growing in logarithmic growth phase in mice right forelimb armpit 0.2ml / (2.5 × 107/ml) inoculation of tumor cells after 6 days, 40 mice were successfully established subcutaneous xenograft model of pancreatic cancer were randomly divided into four groups: normal saline treatment group (control group), 5-FU, PA-MSHA group, PA-MSHA/5-FU group; intraperitoneal injection of saline group 0.2ml physiological intraperitoneal injection of saline ,5-FU group 0.2mg 5-FU (10mg/Kg), Pseudomonas aeruginosa injection (PA-MSHA) group the intraperitoneal the injection 0.2mlPA-MSHA PA-MSHA/5-FU group were injected intraperitoneally 0.2ml PA-MSHA, 0.2mg 5-FU, observed changes in tumor volume and changes in body weight of mice inhibitory rate was calculated, mice were sacrificed after 20 days, eyeball blood, routine measurement of white blood cell count every other day injection time, a total of seven times; ; measure the size and weight of tumor tissue made from the cell suspension line of flow cytometry to detect cell cycle; remaining tumor tissue for pathological confirmation parallel Immunohistochemical detection of P53 protein expression. [Results]: the successful establishment of mouse pancreatic subcutaneous xenograft model; PA-MSHA groups,, PA-MSHA/5-FU group of tumor growth was inhibited, small size, low weight, a statistically compared with the saline group significance (P lt; 0.05); PA-MSHA group, PA-MSHA/5-FU group of white blood cell count was significantly higher than the 5-FU group, the difference was statistically significant (P lt; 0.05); flow cytometry showed that PA- MSHA group, PA-MSHA/5-FU group of cells in G0/G1 phase were higher than the saline group, the difference was statistically significant (P lt; 0.05); immunohistochemistry results are shown: PA-MSHA group, Ⅰ 'A -MSHA/5-FU group P53 protein expression was (43.25 ± 2.62)%, (41.6 ± 4.56)%, significantly lower than the saline group and 5-FU group, the difference was statistically significance (P lt; 0.05) . [Conclusion]: 1 Pseudomonas aeruginosa preparation (PA-MSHA) and PA-MSHA/5-FU respectively enable mouse pancreatic transplantation in tumor volume reduced, weight, tumor growth and proliferation of mouse pancreatic transplantation certain extent; 2 Pseudomonas aeruginosa preparation (PA-MSHA) and] (?) A-MSHA/5-FU respectively of tumor-bearing mice peripheral blood leukocytes had no significant effect; 3 Pseudomonas aeruginosa preparations ( PA-MSHA) can enhance the killing of 5-FU on tumor cells and inhibition: 4 Pseudomonas aeruginosa preparation (PA-MSHA) can inhibit tumor cell proliferation, arrest its evolution from G1 to S phase; 5 Pseudomonas the Pseudomonas preparations (PA-MSHA) and PA-MSHA/5-FU to some extent reduce the P53 positive expression, may have prospects for treatment of pancreatic cancer adjuvant drugs.
