Abstract


Objective:Although the physiological mechanisms contributing to the development of major depression remain unclear, several lines of evidence suggest that the glucocorticoid receptor polymorphism involved in the Pathophysiology and therapeutics to the major depressive disorder. The aim of this study was to investigate the correlation of glucocorticoid receptor gene BclI deletion polymorphism to the major depressive disorder.Methods:We analyzed the GR gene BclIpolymorphism with a polymerase chain reaction (PCR)-based method in 281 patients with major depression and 287 unrelated, age-and sex-matched healthy control subjects. Restriction enzyme BclI was used after PCR amplification to genotype the subjects according to BclI polymorphism of the GR., major depressive disorder was diagnosed according to DSM-IV criteria. In addition, to reduce the clinical heterogeneity, we performed asubtypeanalysis with clinically important variables, such as family history of major affective disorder and age at onset of major depression.Result:. Genotype or allele frequencies is no correlations were detected between major depressive disorder and the control subjects in BclI genotypes freqence,but there is significant difference between the 16 and the 20 years old patients the on the genotype (x2=6.82,df=1,P=0.0330).It is significant difference between the patients which is between 41 and 50 years old patients (genotype:x2=10.696,df=1,P=0.0045) (allele:x2=3.801,df=1,P=0.0161),.The patients with G/C genotype of BclI polymorphism has significant difference between the control when they face to the stress, there is cause factor with them(OR:0.577,P=0.003), and got higher scores on HAM-A compared to patients with G/G and C/C genotypes(P<0.05),(Anxiety(HAM-A<17):OR=2.391,,P=0.003),Anxiety(HAM-A:(18-24):(OR=5.642,P=0.0001) The patients with G/G genotype in MDD is involved in the recurrent episode of MDD(OR=0.678,P=0.035).Conclusion:The result suggest the BclI polymorphism of GR gene more likely be associated with severity of depressive in MDD subjects, and the carrying G/C genotypes patients prone to get higher scores in HAM-A scale, and more sensitive to the stress. There is significant difference between the patients which is between 41 and 50 years old with major depressive disorder and the control subjects in BclI genotypes freqence.There is significant difference was found between 16 and 20 years old.
