Dynamic Monitoring the Relationship among Glutamate, GABA and Dopamine of the Brain in a Rat Model of Parkinson’s Disease

Abstract


Objective: To explore the relationship between glutamate, GABA and dopamine in a rat model of 6-OHDA-induced Parkinson’s disease, so as to study the pathogenesis of Parkinson’s disease.Methods: Brain microdialysis combined with high-performance liquid chromatography (HPLC) was applied to detect dopamine, glutamate, GABA levels in the lesioned striatum at 3-6 weeks post-lesion. At 6 weeks, the rats in these groups were anesthetized, perfused by PBS and fixed by 4% paraformaldehyde (PA) + 0.3% glutaraldehyde. Brains were removed, post-fixed, and made into paraffin sections at 5μm of the substantia nigra, immunofluorescence was used to show TH, Glu and GABA, then which were treated with statistical analysis.Results:(1) Dopamine concentration in PD group decreased markedly post-lesion, which was one-fifth at 5 weeks of Vehicle and Sham group.(2) Contrast to the others, glutamate concentration in PD group was one time higher than the others at 6 weeks post-lesion.(3) GABA concentration at 6 weeks in PD group dropped to one half(4) In PD group, number of GABA+ cell in substantia nigra in the normal side was less than that in the injected side.(5) In the injected side, number of TH+ cells and number of GABA+cell in substantia nigra in PD group were less than these in the other groups; the results in the normal side that PD group was compared to the other groups were same to the injected side.(6) It had shown the double positive cells in the double staining with TH-Rh and Glu-FITC, TH-FITC and GABA-Rh of the striatum.Conclusion:1. Dopamine and glutamate might be mutually reinforcing in the brain of parkinson’s disease rats.2. GABA level in the lesioned side dropped overtime from the onset of parkinson’s disease.3. By some approach the lesioned side and the normal side are interacting, dopamine in the both side, glutamate and GABA in the both side have the same change in the terminal parkinson’s disease.4. The result of immunofluorescence staining suggest that it might have neurotransmitter coexistence in the nerve cells.
