Abstract


Aim of the present study was isolated peripheral blood lymphocytes of RA patients, serum and extract the RNA and protein detection Thl7 cell-specific transcription factors and cytokines related conditions, preliminary study Thl7 cells in rheumatoid arthritis (rheumatoid arthritis, RA) patients and its significance. The first method will be selected based on DAS28 score in RA patients were divided into groups DAS28 ≤ 3.2, 3.2 lt; DAS28 ≤ 5.1 group and DAS28 gt; 5.1 group; were using RT-PCR, Western Blotting Th17 cell-specific transcription factor determination ROR-γt mRNA and protein expression, and Th17 cells by ELISA cytokine levels related to the initial study of Th17 cells in RA patients and its significance. Results 1. Compared with normal control group, RA patients ROR-yt mRNA and protein expression and serum IL-17, IL-21, IL-23 levels were statistically significant (P lt; 0.05). (2) with DAS28 gt; 5.1 group compared to the control group, DAS28 lt; 3.2 group and 3.2 lt; DAS28 lt; 5.1 group ROR-yt mRNA expression were statistically significant (P lt; 0.05); other groups no statistical significance. 3 Compared with normal control group, 3.2 lt; DAS28 lt; 5.1 group and DAS28 gt; 5.1 group of RA patients ROR-yt protein expression were statistically significant (P lt; 0.05); and DAS28 lt; 3.2 set- ratio, 3.2 lt; DAS28 lt; 5.1 group and DAS28 gt; 5.1 group of RA patients ROR-yt protein expression were statistically significant (P lt; 0.05); and 3.2 lt; DAS28 lt; 5.1 group, DAS28 gt ; 5.1 group of RA patients ROR-yt protein expression was statistically significant (P lt; 0.05). 4 Compared with normal control group, 3.2 lt; DAS28 lt; 5.1 group and DAS28 gt; 5.1 group RA patients with IL-17, IL-21, IL-23 serum concentrations were statistically significant (P lt; 0.05 ); and DAS28 lt; 3.2 group, 3.2 lt; DAS28 ≤ 5.1 group and DAS28 gt; 5.1 group RA patients with IL-17, IL-21, IL-23 serum concentrations were statistically significant (P lt; 0.05); and 3.2 lt; DAS28 ≤ 5.1 group, DAS28 gt; 5.1 group of RA patients with IL-17, IL-21, IL-23 serum concentrations were statistically significant (P lt; 0.05). Conclusion 1.RA in PBMC ROR-yt mRNA expression found only in high DAS28 gt; 5.1RA patients. 2.RA ROR-yt in PBMC and serum protein IL-17, IL-21, IL-23 expression seen in a high DAS28 gt; 3.2 in patients with RA. 3.Th17 cells and cytokines in the DAS28 lt; 3.2RA expression levels in patients close to the healthy control group. 4 Tips Thl7 cells may be involved in the disease activity of RA in remission after its expression tended to be normal, as indicators of disease activity as Th17 cells provide an experimental basis.
