Comparison of Role of Several Medicines in Suppressing Bleomycin-induced Pulmonary Fibrosis in Rats

Abstract


Objective To study the dexamethasone (dexamethasone, DXM), N-acetylcysteine ??(N-acetylcysteine, NAC), interferon-gamma (interferon-gamma, IFN-y), low molecular weight heparin (low Molecular weight HEPARIN LMWH), bleomycin (Bleomycin, BLM)-induced pulmonary fibrosis in rats rats. Methods: 150 male SD rats were randomly divided into the healthy control group, pulmonary fibrosis model group, dexamethasone group, NAC intervention group, the the IFN-gamma intervention group and LMWH intervention group, 25 in each group. Each group were 1,3,7,14,28 days were sacrificed 5, collect lung tissue stained by HE and Masson staining to determine lung tissue alveolitis and pulmonary fibrosis, immunohistochemical determination of lung tissue transforming growth factor β1 (transforming growth factor-β1, TGF-β1), collected serum and bronchoalveolar lavage fluid (bronchoalveolar lavage fluid, BALF) line of ELISA for detection of TGF-beta1 content, collected the detection of lung tissue hydroxyproline (hydroxyproline, HYP) content. Results: Compared with model group, the four drugs in the intervention group alveolitis and pulmonary fibrosis at day 28 were significantly reduced (P lt; 0.05); comparison between the four drug intervention group, alveolitis and pulmonary fiber degree in 28 days no significant difference (P gt; 0.05); 2. Compared with model group, the four drugs in the intervention group lung tissue TGF-beta1 expression levels in 14 and 28 days was significantly lower (P lt; 0.05 ), NAC group, the the LMWH group with dexamethasone group lung tissue TGF-beta1 expression levels (28 days) was significantly lower (P lt; 0.05); NAC group LMWH group, IFN-gamma group between lung organization of the expression level of TGF-beta1 was no significant difference (P gt; 0.05); four drug intervention group compared with the model group, serum and BALF in the expression level of TGF-beta1 on the 7th, 14th, 28th day lower (P lt; 0.05), which NAC group (14, 28 days), LMWH group (14 days) than in the dexamethasone group (P lt; 0.05); NAC group, LMWH group, IFN-gamma between the two groups in the serum and BALF TGF-beta1 expression level was no significant difference (P gt; 0.05); 4. HYP content of the four drugs in the intervention group compared with the model group was significantly reduced in the 7th, 14th, 28th day (P lt; 0.05), which the NAC group (14, 28 days), LMWH group (28 days) than the dexamethasone group more obvious (P lt; 0.05); the NAC group, LMWH group, IFN-gamma group between the HYP content was no significant difference (P gt; 0.05) Conclusion: In normal lung tissue, TGF-beta, a small amount of expression of TGF-beta1 expression increased significantly in the model of bleomycin-induced pulmonary fibrosis in rats. Dexamethasone, N-acetyl-cysteine, and interferon-gamma low molecular weight heparin can reduce bleomycin-induced pulmonary fibrosis in rats. The 3.N-acetylcysteine ??and low molecular weight heparin improvement of pulmonary fibrosis than dexamethasone. 4 of interferon-gamma on the degree of improvement of pulmonary fibrosis and dexamethasone compared to no significant difference. 5 of dexamethasone, N-acetylcysteine, interferon-gamma, low molecular weight heparin anti-fibrosis and to reduce the expression of TGF-beta1.
