Abstract


Purpose: blood stasis and promoting blood soup is the national old TCM Sun Tong Jiao professor in the practice of long-term clinical summary of out of the treatment of non-alcoholic fatty liver disease experience side, is the \\Both to the theory of traditional Chinese medicine as a guide, and a combination of modern pharmacological research, significant efficacy in clinical applications. The purpose of this project is to clarify the pharmacological effects of blood stasis and promoting blood soup, to explore its mechanism, to provide a scientific basis for clinical treatment, and lay a good experimental basis for further research. Methods: 60 male SD rats were randomly divided into 2 groups: normal control group (10), one week after the feeding under experimental conditions, given a normal diet; model group (50), given the high-fat diet. In the second week after the high fat diet, the model group, 50 rats were divided into model control group, low-dose treatment group, medium-dose treatment group, high-dose treatment group, tiopronin treatment group. Low Compound expectorant and blood soup Chinese medicine in the high dose group were given a dose of 0.5 times (6g/kg.d) times the dose (12g/kg.d), 1.5-dose (16g/kg.d). Tiopronin treatment group to give drugs 40mg/kg.d. Model control group, normal control group without any drugs, and replace them with the same dose of saline gavage. Experimental animals free water and eating, weighed every 2 weeks, and adjust the dosage accordingly. Packet dosing regimen in accordance with the above, 8,9,10 weekend, each from the model group were randomly selected 1-2 rats were sacrificed to observe the pathological changes of the liver tissue, and blood biochemical parameters (ALT, AST, TG, TC, etc.). 10 weeks, all rats were sacrificed and liver function, blood lipids measured the same way, the liver tissue with 10% formaldehyde solution fixed, paraffin-embedded serial sections, slice thickness of 5μ m HE staining and TUNEL assay cells wither perish. Results: The model group and normal control group, their body weight, liver weight and serum ALT, AST, TC, TG, liver histopathology degree of inflammatory activity, the degree of fatty degeneration of the liver cells, the expression of hepatocyte apoptosis index significantly elevated heavier (P lt; 0.01); low dose treatment group, medium-dose treatment group, high-dose treatment group, the of tiopronin treatment group and model group comparison, the indicators are significantly reduced or mitigated (P lt lower body weight of rats of the low dose group compared with the model control group showed no significant difference (P gt; 0.05); higher body weight between the dose of Chinese medicine in rats showed no statistical significance (P gt;; 0.05); 0.05); tiopronin treatment group and high dose traditional Chinese medicine group between liver weight contrast, no difference in the data. (P gt; 0.05); liver weight of the low dose group in the dose of Chinese medicine group difference was not statistically significant (P gt; 0.05); Chinese medicine of high, medium and low-dose group, tiopronin treatment group group within the data, ALT, AST change no differences (P gt; 0.05); expectorant promoting blood soup low-dose group and the model control group, serum TG, TC no difference (P gt; 0.05); of tiopronin treatment group and the high dose treatment group inflammatory activity score distinction is not obvious, no statistical significance (P gt; 0.05); fatty degeneration of the liver cells of the low dose group higher than the of tiopronin treatment group and the high dose group (P lt; 0.01) and middle dose of Chinese medicine group, steatosis differences (P lt; 0.05); of tiopronin treatment group and the high dose group no difference between degree of fatty degeneration of liver cells (P gt; 0.05); expression of hepatocyte apoptosis index in the high dose group, between the tiopronin treatment group showed no significant difference (P gt; 0.05); Conclusion: expectorant Huoxue soup can significantly alleviate non-alcoholic fatty liver (NAFL) the degree of fatty degeneration of the liver tissue of rats, regulate lipid metabolism disorders, to some extent, improve liver function, reduce apoptosis index, which may be its prevention and treatment of non-alcoholic fatty liver disease ( NAFLD), one of the mechanisms.
