Abstract


Objective: To investigate Chinese medicine amethyst in anovulatory rat model of ovarian local INH , IGF - I affect the mechanism of action and amethyst pharmacological effects of the material basis , to provide a theoretical basis for the amethyst in the clinical treatment of ovulatory disorder infertility . Methods: minors 24d old SD female rats 60 , weight 75 ± 5g were randomly divided into model group , clomiphene group , kidney blood circulation parties group , amethyst group , the normal group , calcium fluoride group , ( n = 10 ) . Bogovich method to establish ovulation disorders pathological model in rats . Kidney blood circulation parties and clomiphene citrate ( CC ) for the control rat model of ovarian ovulation disorders observed by immunohistochemistry amethyst local inhibin ( INH ) , insulin-like growth factor - I ( IGF - I ) protein expression ; and amethyst group compared with the experimental results of the calcium fluoride group to analyze the possible material basis of amethyst corresponding pharmacological effects . Results: Compared with normal group , ovarian local tissue IGF-I with INH expression intensity was abnormally high expression , the difference was significant ( P lt ; 0.01 ) . Amethyst , kidney blood circulation parties , clomiphene citrate and calcium fluoride treatment , ovarian local tissue IGF-I with INH protein expression were significantly lower ( P lt ; 0.01 , P lt ; 0.05 ) , the amethyst , kidney and Activating Blood square , clomiphene citrate and calcium fluoride inhibit expression of IGF-I with INH . Amethyst , kidney blood circulation parties group and clomiphene group pairwise comparison , the difference was not statistically significant ( P gt ; 0.05 ) . Amethyst rats ovarian local IGF - I with INH average optical density values ??than those of calcium fluoride group to be small, but the difference was not statistically significant . Conclusion : IGF - I with INH may be involved in the pathogenesis of ovulation disorders . The amethyst can regulate ovulation disorders abnormally high expression of the the rat ovary local IGF - I with INH and the results are slightly better than calcium fluoride . Prompt the regulatory function abnormally high expression regulating the local ovarian IGF-I with INH , amethyst compared with the role of calcium fluoride ; amethyst may have hormone - like effect , enabling the hypothalamus - pituitary - ovarian axis ( HPOA ) can be improved, so as to promote primary follicles to the dominant follicle development , but its corresponding material basis for further study .
