Effects of Compound Traditional Chinese Medicine on Immunologic Function in Mice

Abstract


The object of this research is to make sure of effects of compound traditional chinese medicine on the immunological function in mice, in order to develop domestic animal tranditional chinese medicine immunopoteniator finally. Compound traditional chinese medicine, involving Astragalus, Angelica Sinensis and Prepared Licorice and so on, was decocted by water and compound liquid medicine was obtained. Security of compound liquid medicine was detected by mice’s acute toxicity tests (median lethal dose test and maximum tolerance test).After immunodepression of pathological model mice were got by using cyclophosphamide, these mice and other healthy mice were divided into four groups simultaneously, including three different concentration treatment groups and one control group. The concentration of three different treatment groups were 1.0g/mL, 0.5g/mL, 0.25g/mL respectively, and the water of one control group was 0.5mL/d, and then fifteen days later, hematological indices which included red blood cell, white blood cell, hemoglobin content, hematocrit, neutrophil, mononuclear cells, lymphocyte, eosinophils and basophils, spleen visceral index and body weight increase were tested. After that, relation between concentration and efficacy of compound traditional chinese medicine was studied, in the end, the best concentration of compound traditional chinese medicine was confirmed. In order to make sure of effects of compound traditional chinese medicine on the immunological function in mice, this research, by using cytometry, colorimetric method and rachel’s dyeing microscopy method, has studied compound traditional chinese medicine on T achroacyte subgroup(CD4~+, CD8~+), B lymphocytes serum hemolysin (IgM), Abdominal macrophage phagocytosis function (Phagocytic rate, phagocytic index) in healthy mice and in immunodepression mice.This research showed the median lethal dose test of compound traditional chinese medicine was >50g/kg·BW, and the maximum tolerance test of compound traditional chinese medicine was >150g/kg·BW. In two tests between compound traditional chinese medicine group and control group, compared with the control group, the compound traditional chinese medicine group of hematological indices which included red blood cell, white blood cell, lymphocyte, spleen visceral index and body weight increase were all increased and presented concentration - efficacy positive correlation. In the tests of healthy mice compound traditional chinese medicine group, compared with the control group, the difference between number of CD4~+/CD8~+ was not significant (P>0.05), but the difference of IgM and devouring percentage were significant (P<0.05), the difference of phagocytic index was extremely significant(P<0.01). In the tests of immunodepression mice compound traditional chinese medicine group, compared with the control group, the difference of number of CD4~+/CD8~+, IgM, devouring percentage and phagocytic index extremely significant were all extremely significant (P<0.01).In sum, this research showed this kind of compound traditional chinese medicine was security, and when the concentration was 1.0g/mL, the efficacy is the best, which had the function of enhancing immune system of mouse, especially, this kind of compound traditional chinese medicine still had more immunopotentiation in immunodepression mice.
