Abstract

Sarcopenia is the skeletal muscle degenerative changes occur with the aging process, resulting in the lowering of the elderly muscle strength, muscle reduction, reduced balance capacity, fatigue, frequent falls, even causing a fracture, a heavy burden to families and society . Many experts and scholars in their tireless efforts to explore Sarcopenia pathogenesis is widely accepted that calcium homeostasis changes with the the Sarcopenia occurrence is closely related to calcium homeostasis in the skeletal muscle of aging changes can directly reduce the lead to muscle function and muscle loss. MG29 protein is a transmembrane protein of the newly discovered mechanism for the regulation of calcium, the study found that the MG29 protein in the aging rat skeletal muscle reduced by about 50% compared with normal young rats. Was the normal young mouse the MG29 protein gene knockout and MG29 missing young rat skeletal muscle appears similar to the characteristics of aging rats. Predecessors many studies have confirmed Sarcopenia muscle function in patients with trainable, especially in the resistance movement effect is more significant. However, whether the correlation between the movement and MG29 protein? If there is, its relevance and how? Currently a lack of relevant reports. Explore the relationship between the MG29, resistance exercise and Sarcopenia, which explain the mechanism of muscle lost their trainability, this study, 32 16-month-old male Wistar rats were randomly divided into four groups, for the control group (CG), and the remaining three groups for the exercise group were larger load load running sports group (HG), medium load weight-bearing running exercise group (MG) and a smaller load weight run sports group (LG) of CG group does not impose any exercise intervention the mouse exercise group to be eight-week weight-bearing treadmill training, treadmill running speed 15m/min gradient of 0 ° taken ran 2mim, the rest 2mim for a group of six groups, continuous training, once daily, closed on Sundays. Weighed weekly, and observe the change of coat color, mental state appearance. Rat Maximum weight percentage of the weight of the load to the the larger load negative reorganization negative weight equivalent to 60% of the maximum load, medium load negative reorganization weight equivalent to 50% of the maximum load, smaller line with negative recombinant weight equivalent to 30% of its maximum load. In the eighth week of the experiment, after the last movement of materials, take each rat left rectus femoris production of conventional frozen sections, HE staining using an optical microscope to observe the general form of the muscle tissue, and then were to take the right amount of muscle tissue homogenates using fluorescence colorimetric assay its lipofuscin methylthymol blue colorimetric detection of calcium concentration and determination MG29 protein expression using Western blot (Western blot). Software Excel 2003 and SPSS17.0 summary data and statistical analysis, comparing the results within each group and between groups, concluded. Results: (1) training, the general state of mind of the movement of the rats was significantly better than the CG group. Longitudinal analysis of the the weight exercise group, and was decreased and then increased, then decreased trend. And the the CG group's weight there is a steady increase in the trend. (2) light microscope to observe the morphology of the muscle cell samples round of the muscle fibers, with varying degrees of sizes, cell malalignment, the gap is larger, and the middle of white tissue filling nuclei showed centralized. The most obvious CG group HG group, followed by LG and MG group of muscle fibers more closely than the other two groups of permutations. (3) The homogenates detection of lipofuscin concentration, the results show the LG group and MG group does not have a significant difference, the content thereof is less; HG group and the the CG group not having a significant difference, the higher the content thereof, significantly higher than LG and the MG group (P lt; 0.05). (4) homogenate detect calcium concentration, results showed that the highest content of LG group, there is a significant difference compared to the other groups, no significant difference between MG and HG groups CG group content of at least, compared to the other groups also have significant differences. (5) Western Blot Detection homogenate MG29 protein expression levels, analyze electrophoresis pattern showed that the highest concentrations of the LG group, there is a significant difference compared to the other groups, followed by MG group, again the CG group, the least HG group content, and more Other groups have significant differences. Conclusions: (1) the appropriate intensity intermittent load run training can effectively improve the the MG29 protein levels within the aging rat skeletal muscle cells, effectively improve the morphology and function of skeletal muscle cells, thereby delay to some extent due to the increasing age muscle atrophy, loss, slow down the process of Sarcopenia. 30% and 50% of the maximum load intermittent load run exercise may be more appropriate to delay the aging of rat skeletal muscle motor stimulation, which can effectively improve the the MG29 protein expression in the skeletal muscle level, so that the skeletal muscle to maintain normal the excitation-contraction coupling structure and promote the inflow of extracellular calcium, to maintain or enhance the normal contractile function of skeletal muscle, relatively lower levels of free radicals in the cells, thus delaying muscle atrophy, loss caused due to aging, slow down the process of Sarcopenia. (2) 60% of the maximum load intermittent load running exercise on aging skeletal muscle may be inappropriate motor stimulation, but to make due to the intensity movement MG29 protein in rats reduced the contractile function of skeletal muscle weakening, and The relative radical levels in skeletal muscle, a large number of lipofuscin accumulation. The intensity exercise may accelerate the aging process in skeletal muscle.
