First, the study of several different surface graft chain length of polystyrene PAA - polyacrylic acid (PS-PAA) spherical polyelectrolyte brush in phosphate buffered saline (PBS) in the bovine serum albumin (BSA) physical adsorption, PAA obtained graft chain length of PS-PAA spherical polyelectrolyte brush physical adsorption of BSA were studied: in a certain chain length (25nm), the length of the graft chain with the increase of PAA, PS-PAA spherical polyelectrolyte brush of BSA reduced physical adsorption; when PAA graft chain length to a certain extent (25nm) is, PS-PAA spherical polyelectrolyte brush physical adsorption of BSA reaches a minimum value 33μg/mg, the graft chain length continues to increase, PS- PAA physical adsorption of BSA does not change. The above results for the PS-PAA spherical polyelectrolyte brushes used in the surface structure of the real physiological environment choose to provide preliminary reference. Secondly, the study of the surface graft chain length different PAA PS-PAA spherical polyelectrolyte brush in PBS and biotin for streptavidin (SA) Adsorption of the chemical. In PBS buffer, by N-hydroxysuccinimide (NHS) and carbodiimide (EDC) activation of PS-PAA PAA spherical polyelectrolyte brush chains on the carboxyl, activated carboxyl group of the amino group with SA forming a chemical bond to achieve SA chemisorption. Then through the acid-base titration to determine the conductivity of different PAA graft chain length PS-PAA spherical polyelectrolyte brushes carboxyl content on the particles, the final analysis of the carboxyl content and PAA graft chain length on the PS-PAA spherical polyelectrolyte brushes in PBS SA buffer chemisorption of law: With PAA graft chain length and a carboxyl content increases, PS-PAA spherical polyelectrolyte brush the SA of the chemical adsorption capacity gradually increased, and the higher the carboxyl content, PS-PAA spherical polyelectrolyte brush the SA chemisorption ability to increase faster. The results of the above study biological applications PS-PAA spherical polyelectrolyte brush structure designed to provide a foundation theory. Finally, a preliminary study of the PS-PAA spherical polyelectrolyte brushes chemisorbed SA and freedom biotin (biotin) binding activity. First through competitive inhibition of the enzyme assay when the PS-PAA spherical polyelectrolyte brushes in the SA density is low, PAA graft chain length on the biotin binding activity of SA, SA showed low density, PAA graft chain length on SA biological activity had no significant effect; then further testing when the PAA graft chain length is shorter (12.5nm) when, PS-PAA spherical polyelectrolyte brushes in the density of the SA SA biotin-binding activity of the law, the results showed that: when the PAA graft chain length is short (12.5nm), PS-PAA spherical polyelectrolyte brushes in the SA SA density and freedom of biotin-binding activity of no significant impact. This section chemisorption SA, PS-PAA used in biomedical sensing applications provide some reference value.
