Abstract


Rice (Oryza sativa L.) as one of the most important food crops, with the population growth and the reduction of arable land, improve rice yields for solving the problem of food security in the future has a very important strategic significance. Rice grain weight three elements constitute the rice yield (panicle, grain number, grain weight) one. Grain weight is affected by multiple quantitative trait loci (QTL) controlling a complex trait that the majority of studies for grain weight is limited to use primary mapping populations for QTL analysis. In this study, grain weight have significant differences in rice varieties \), based on the constructed mapping populations; to the length and width of grain trait research, conducted a preliminary positioning, fine positioning and highly accurate linkage analysis. Eventually we fine positioning control of rice grain length and grain weight QTL GW6, and try the big grain gene GW6 with GIF1 (grain incomplete filling 1) gene pyramiding, and build introgression lines will GW6 gene into 9311 to get a good intermediate materials. The main results are as follows: 1, GW6 backcross population construction: the GS3 molecular markers tightly linked to the exclusion of the two near-isogenic lines long grain effect GS3 gene was constructed GW6 backcross population, for us to be studied separately GW6 gene function; 2, GW6 classical genetic analysis: examine BC 3 F 1:2 , BC 4 F 1:2 , and F 1:2 various groups of grain length / grain weight showed that GW6 an incomplete dominant gene, not the results of previous studies indicate GW6 recessive gene; 3, GW6 fine positioning: use BC 3 F 2 , BC 4 F 2 , NIL-GW6 GW6 fine mapping of the gene, will GW6 positioned at 200 kb range, combined with bioinformatics prediction of possible candidate genes GW6 gene - OJ1001 B 06.34; 4, GW6 gene prediction: Determination OJ1001 B 06.34 whole genome sequences and found a large grain germplasm treasure large grains in the gene second exon deletion of 13 bp nucleotide and protein sequence analysis where nucleotide deletion causes premature termination of protein translation; 5, GW6 relevant tag design : According to the sequencing results designed OJ1001 B 06.34 gene function markers GW6-1, to facilitate this gene pyramiding of use; 6, molecular breeding: Flower 11 in both genetic background, building NIL -GW6 will import a large grain gene GW6 flower 11 flower 11 significantly increased the grain size, improve grain weight; introgression lines will try to build GW6 gene into the middle of 9311 was a good material.
