Abstract


Objective: endometrial cancer (endometrial cancer EC) is one of the three major malignancies of the female genital tract, the current incidence of an upward trend in the world. Cyclooxygenase -2 (COX-2) is overexpressed in a variety of tumor tissue, suggesting that its participation in the development of tumor. Combining the corresponding receptor in tumor cells, the signal transduction pathway is through its downstream product to complete, while one of its important receptor is a peroxisome proliferator activated receptor-gamma (PPAR-gamma). In recent years, the study found that PPAR-γ is the intersection of multiple signaling pathways of the body, can induce differentiation of tumor cells. PPAR-γ and obesity, fat metabolism and insulin resistance are closely related, there is a certain relationship prompted with the development of endometrial cancer. In summary, we speculate the COX-2mRNA, PPAR-γmRNA may endometrial cancer showed high expression by RT-PCR assay both endometrial cancer and normal endometrial tissue expression and related and further analysis of the relationship of both with the type of endometrial cancer, clinical stage, histological grade and other clinicopathological parameters, provides new clues for the diagnosis and treatment of endometrial cancer. Method: Select the Second Affiliated Hospital of Hebei Medical and Fourth Affiliated Hospital of Obstetrics and Gynecology, 28 cases of endometrial cancer underwent surgery in September 2009 to December 2010, to take a full cut of endometrial carcinoma; take 20 cases of normal endometrial tissue over the same period total hysterectomy due to uterine benign line. All cases were confirmed by pathology. Endometrial cancer staging by the International Federation of Gynecology and Obstetrics (FIGO, 2009), surgical - pathologic staging criteria, all objects in the collection of samples did not receive radiotherapy, chemotherapy or hormone therapy. Results: 1 endometrial carcinoma and normal endometrial tissue in both COX-2, PPAR-γ expression, but in the endometrial carcinoma the COX-2mRNA, PPAR-γmRNA expression was significantly higher than the normal uterus endometrial tissue, the difference was statistically significant (P lt; 0.05). 2 COX-2mRNA the expression of PPAR-γmRNA the organization with lower degree of differentiation and increased, and the difference was statistically significant (P lt; 0.05). The expression of COX-2mRNA cancer tissue myometrial invasion depth of myometrial invasion gt; 1/2 expression was significantly higher than that of myometrial invasion ≤ 1/2, the difference was statistically significant (P lt; 0.05). Of PPAR-γmRNA the expression and endometrial cancer FIGO surgical - pathologic staging was positively correlated, the difference was statistically significant (P lt; 0.05). Whether expression of COX-2mRNA, PPAR-γmRNA endometrial cancer tissue type and lymph node metastasis, the difference was not statistically significance (P gt; 0.05). 3 endometrial cancer tissue of COX-2mRNA the expression of PPAR-γmRNA the expression was positively correlated (r = 0.624, P lt; 0.05). Conclusions: 1 of COX-2 mRNA expression of PPAR-γmRNA in endometrial cancer significantly Ming higher than in normal endometrium and endometrial cancer of COX-2mRNA the PPAR-γmRNA the expression was positively correlated, suggesting that two play a collaborative role in the occurrence and development of endometrial cancer process. 2 of COX-2 mRNA, the level of of PPAR-γmRNA expression amount and prognosis of endometrial cancer is closely related of COX-2 mRNA the PPAR-γmRNA expression in patients with endometrial cancer tissue differentiation is poor, the high degree of malignancy, clinical stage late and poor prognosis. Combined detection of COX-2, PPAR-γ is expected to become important indicators predict the development of endometrial cancer and prognosis.
