Expression of TGF-β2, PR, MMP-3 and MMP-7 and Their Significance in Eutopic Endometrium with Endometriosis in Mice

Abstract


Objective:The morbidity rate of EMs gradually increased in recent years,but the occurrence of the mechanism has not been completely set. Recent research showed that the .pathogenesis of EMs was related to TCDD. Amounts of researches indicated that TCDD promoted the occurrence and development of EMs. But the mechanisms which promote the occurrence and development of EMs have not been clarified. Research showed that dioxin probablly affected the biological function of eutopic endometrium of the cause of EMs. And prenatal exposure may increase the risk of disease in adulthood. The prolonged exposure of TCDD can promote the development of the EMs. Some people reported that TGF-β2,PR,MMP-3 and MMP-7 play an important role in the normal functions ofendometrium. This experiment tested the expression of TGF-β2,PR,MMP-3 and MMP-7 in eutopic endometrium of the model of EMs in mice for exploreing the nosogenesis of EMs.Methods:1 Establishment of Animal ModelsSelecting 60 pregenant SPF C57BL/6 mice ,developmental exposure of mice to TCDD to their femal offsprings, and divided into five groups:0/0,0/3 , 3/0 , 3/3 , 3/10 ( control , puberty+adulthood , fetus period+ puberty+adulthood,high dose exposure).The mouse endometriosis model was established with mouse autotransplantation of endometrium at the seventieth day after birth,after surgery the treatment groups were exposured to 3ug/kg TCDD every three weeks and euthanized at 12 weeks after surgery.2 Detection of IndicatorsAfter obtaining eutopic endometrium tissues,the expression of TGF-β2,PR,MMP-3 and MMP-7 were detected by immunohistochemical and Western blotting methods and compared the data.3 All data were analyzed withSPSS13. 0 statistical software package. Measurement data was noted by formulas of x±s and analysis them by one-way ANOVA. Enumerationdata was analyzed by chi-square test. P<0.05 was considered significant for all statistical analysis.Results:1 Comparison of success ratio about the EMs model in mice. The data was analyzed by chi-square test. The significant different data between groups was analyzed by multiple comparison .Result showed that the success ratio of ectopic plant in hypodermic between different groups was not significant different.( P>0.05).The success ratio of ectopic plant in peritoneum was significant different(P<0.05).The result of pairwise comparison showed that the success ratio of 3/3 was significant higher than that of 0/0(P<0.00625).The success ratio of ectopic plant in ovary was not significant different between groups(P>0.05).The success ratio of ectopic plant in hypodermic, peritoneum and ovary was not significant different within 0/0(P>0.05).The success ratio of ectopic plant between different position within 0/3 was significant different(P<0.05).The result of pairwise comparison showed that the success ratio of in hypodermic and peritoneum or in peritoneum and ovary was not significant different(P>0.0125, P<0.0125).The success ratio of ectopic plant in ovary was significant higher than that in hypodermic(P<0.0125).The success ratio of ectopic plant in hypodermic, peritoneum and ovary was significant different within3/0(P<0.05),The result of pairwise comparison showed that the success ratio of in hypodermic and peritoneum or in peritoneum and ovary was not significant differen(tP>0.0125, P>0.0125).The success ratio of ectopic plant in ovary was significant higher than that in hypodermic(P<0.0125).The success ratio of ectopic plant in hypodermic, peritoneum and ovary was significant different within3/3(P<0.05).The result of pairwise comparison showed that the success ratio of in hypodermic and peritoneum or in peritoneum and ovary was not significant different ( P>0.0125 , P>0.0125).The success ratio of ectopic plant in ovary was significant higher than that in hypodermic(P<0.0125).The success ratio of ectopic plant in hypodermic, peritoneum and ovary was significant different within3/10(P<0.05).The result of pairwise comparison showed that the success ratio of in hypodermic and peritoneum or in peritoneum and ovary was not significant different(P>0.0125,P>0.0125).The success ratio of ectopic plant in ovary was significant higher than that in hypodermic(P<0.0125).Research also found that aspect of ectopic focus developed from small vesicle to big vesicle following the addness of dosage and large amounts of fluid was accumulated in it,and the surface tension of the vesicle was very big and abdominal cavity synechiaed severely.2 The expression of TGF-β2,PR of eutopic endometrium in mice between different groups.The expression of TGF-β2 and PR between 0/0 and 0/3 was not significant different( P>0.05,P>0.05).The expression of which between 0/0 and 3/0 was not significant different(P>0.05,P>0.05). The expression of TGF-β2 or PR in 0/0 was significant lower than 3/3 and 3/10 (P<0.05,P<0.05). The expression of which in 3/3 was significant more different than that in 3/10(P<0.05).The result of western-blot verified that of immunolhistochemistry by quantification analyzed.3 The expression of MMP-3 and MMP-7 of eutopic endometrium in mice between different groups.The expression of MMP-3 and MMP-7 between 0/0 and 0/3 was not significant differen(tP>0.05,P>0.05).The expression of MMP-3 and MMP-7 between 0/0 and 3/0 was not significant different(P>0.05,P>0.05).The expression of MMP-3or MMP-7in 0/0 was significant lower than 3/3 and 3/10(P<0.05,P<0.05)The expression of which in 3/10 was significant higher than in 3/3(P<0.05).The result of western-blot verified that of immunolhistochemistry by quantification analyzed. Conclusions:1 After ectopic plantation in hypodermic,peritoneum and ovary of mice which in different times and different dose of TCDD exposure ,the success ratio of ectopic plant in peritoneum of 3/3(fetus+ puberty+adulthood)was significant higher than that of 0/0(control) .In different dose of TCDD exposure groups ,the success ratio of ectopic plant in ovary was significant higher than that in hypodermic compare to control group. Those noted that TCDD exposure can promote the plantation of ectopic endometrium.2 The expression of TGF-β2 and PR in eutopic endometrium of 3/10 (fetus + puberty+adulthood high dose of TCDDexposure group )mice was significant lower than that of 3/3 (fetus + puberty+adulthood ordinary dose of TCDDexposure group),and the expression of TGF-β2 and PR in that of 3/3 was significant lower than that of 0/0 , Thus concluding that TCDD down-regulated the expression of TGF-β2 and PR and the intensive function of TCDD was increased following the added dosage.3 The expression of MMP-3and MMP-7 in eutopic endometrium of 3/10 (fetus + puberty+adulthood high dose of TCDD exposure group )mice was significant lower than that of 3/3 (fetus + puberty+adulthood ordinary dose of TCDDexposure group),and the expression of MMP-3 and MMP-7 in that of 3/3 was significant lower than that of 0/0,Thus concluding that TCDD down-regulated the expression of MMP-3 and MMP-7and the intensive function of TCDD was increased following the added dosage.4 Different dose of TCDD exposure to fetus、puberty and adulthood stage mice promoted the occurrence of EMs in adulthood through altering the physiological functions of eutopic endometrium .
