Objective: To observe the immunomodulatory and anti- angiogenic effect of thalidomide on experimental rats with ulcerative colon whether therapeutic effect . Methods: 40 SD rats were divided into normal control group (CON group , n = 10) and experimental group (n = 30), experimental rats through a free drink 3% dextran sulfate sodium (DSS) was established chronic ulcers colitis model , and then divided into experimental group (M group , n = 10), the early treatment group (S1 group , n = 10, 8-21 days to give thalidomide ) , late treatment group ( S2 group , n = 10, the first 25-38 days given thalidomide ) , observed : rat disease activity index (DAI), colon tissue damage index (HI), colonic mucosa mean vessel density (MVD) and intravascular micro- thrombosis , and serum TNF-α, VEGF concentration . Results: Compared with CON group HI (1.9 ± 1.2), MVD (61.1 ± 9.8) compared , M group HI (6.1 ± 2.8, q = 6.54, P lt; 0.05), MVD (114.0 ± 5.0, q = 13.56, P lt; 0.05) were significantly increased , but the serum TNF-α levels (CON group 265.4 ± 56.8 vs M group 254.4 ± 90.8, q = 0.61, P gt; 0.05), VEGF levels (CON group 182.8 ± 19.3 vs M group 194.7 ± 34.9, F = 2.671, v1 = 3, v2 = 34, P = 0.063 gt; 0.05) difference was not statistically significant. S1 group (2.7 ± 1.6, q = 4.24, P lt; 0.05), S2 group (3.0 ± 1.7, q = 3.60, P lt; 0.05) DAI than M group (4.7 ± 1.8), but with the HI group M (6.1 ± 2.8) compared to the group S1 HI (6.0 ± 1.7, q = 0.17, P gt; 0.05) and S2 group HI (5.8 ± 1.9, q = 0.48, P gt; 0.05) did not significantly decreased ; 4 groups of animals serum VEGF levels showed no significant difference (CON: 182.8 ± 19.3, M: 194.8 ± 34.9, S1: 169.7 ± 13.6, S2: 169.7 ± 15.4, F = 2.671, v1 = 3, v2 = 34, P = 0.063 gt; 0.05 ) ; and M serum TNF-α levels (254.4 ± 90.8) compared to , S1组(159.3 ± 20.7, q = 4.96, P lt; 0.05), S2组(172.1 ± 23.4, q = 4.55, P lt; 0.05 ) reduction in the level of serum TNF-α , also , S1 group MVD (89.8 ± 15.1, q = 6.05, P lt; 0.05), S2 group MVD (92.6 ± 13.6, q = 5.35, P lt; 0.05) is lower than M group (114.0 ± 5.0). Group 4 intravascular microthrombi positive rate was no significant difference (CON: 30.0%, M: 33.3%, S1: 66.7%, S2: 60.0%, χ2 = 3.982, v = 3, P ≈ 0.263 gt; 0.05) . Conclusion: Thalidomide has anti- TNF-α and anti- angiogenic effect, can reduce disease activity index UC rats , does not increase the risk of blood clots forming locally , but the UC histological improvement seems to be no apparent effect .
