Abstract


Objective: Diabetic fatty liver (Diabetic Fatty Liver, DFL) refers to diabetes (Diabetes Mellitus, DM) based on the emergence of fatty degeneration of liver cells, which can be clinically attributed to non-alcoholic fatty liver disease (Nonalcoholic Fatty Liver Disease, NAFLD) category. \kind of damage factor is the second blow, causing further liver degeneration, inflammation, necrosis and fibrosis. Second hit in the pathogenesis of DFL was particularly prominent. This study of SD rats were injected intraperitoneally in high-fat diet combined dose of streptozotocin (Streptozotocin, STZ) method to establish DFL model, application Yiqiyangyin phlegm meridians traditional Chinese medicine intervention of the model by observed experimental animals generally, liver index, liver pathological, serum triglyceride (TG), cholesterol (TC), high density lipoprotein (HDL) levels, liver tissue malondialdehyde (MDA) levels, total liver tissue superoxide dismutase (T-SOD) activity, serum adiponectin levels, liver tissue adiponectin (Adiponectin) and its receptor 2 (Adiponectin Receptor2, AdiR2) levels to explore phlegm meridians nourishing herbal compound the DFL therapeutic effect and its possible mechanism of action for the clinical treatment with Qi phlegm network as the basic pathogenesis of diabetes and provide experimental evidence. Method: Clean the grade 75 healthy male SD rats, weighing 240 2 60g, adaptive feeding one week later, were randomly divided into normal group 12, model 18, Yiqiyangyin phlegm meridians low dose group, middle dose group and high dose group in each group 15. In addition to the normal group was given normal diet, the rest of the rats were given high fat diet. Feeding four weeks, in addition to the normal group, the rest of the group were a single intraperitoneal injection of STZ 35mg · kg -1 , the normal group were injected considerable dose citrate buffer, and 72 hours after three consecutive second tail vein blood glucose measurements to ≥ 16.7mmol · L -1 of DM model modeling success criteria. After the modeling, phlegm meridians of Chinese medicine Yiqiyangyin low, medium and high dose groups were given medicine 5.29g · kg -1 · d -1 , 10.58g · kg -1 · d -1 , 21.16g · kg -1 · d -1 gavage, the normal group and model group were given equivalent doses of saline once daily for 8 weeks. In the first 8 weeks after treatment, fasting 12h, weighed under anesthesia, the femoral artery blood serum was separated, measured TG, TC, HDL, ELISA was used to detect serum Adiponectin levels; removal of the liver, liver weighed liver weight index of liver tissue specimens placed in 4% paraformaldehyde, rats were observed under light microscope morphological changes in liver pathology; liver tissue specimens frozen in liquid nitrogen cryogenic rear -80 ℃ refrigerator for detecting liver tissue of MDA, T-SOD activity, RT-PCR method for detection of liver tissue Adiponectin levels, Western blot was used to detect the expression of liver AdiR2. Result: a general condition of rats during the experiment, in addition to the normal group, the other four groups were death cases in which the model group died 4, Chinese high-dose group died 2, middle and low dose group died each one, The cause of death may be associated with ketoacidosis caused by high blood sugar and various organ disorders. Normal rats showed a progressive increase in weight, eating properly, responsive vision of God, shiny hair, feces were grain samples, urine normal. The rest of the model group rats by intraperitoneal injection of STZ, there was obvious polydipsia and polyuria phenomenon, and the gradual emergence apathetic, unresponsive, eyes dull, dull hair, feces and sometimes shapeless. Two rats in each group body weight, liver weight, liver index (liver weight / body weight × 100%) results showed that: the rats body weight and liver weight, compared with the normal group, model group, low, medium, high dose group were body weight, liver weight was significantly higher (P lt; 0.05); Compared with model group, Chinese medicine each dose group body weight, liver weight was significantly lower (P lt; 0.05); between each treatment group without significant difference (P gt; 0.05); each rat liver index, compared with the normal group, model group, low, medium and high dose group were significantly elevated liver index (P lt; 0.05); and Model group, Chinese medicine liver index each dose group was significantly lower (P lt; 0.05); each treatment group was no significant difference between (P gt; 0.05). Tips Yiqiyangyin phlegm meridians of Chinese medicine can significantly reduce the DFL rat body weight, liver weight and liver index, reduce liver steatosis. Three rats in each group pathological changes in the liver lobule normal rats complete, clear boundaries, liver cells arranged radially around the central vein, the central large round nucleus, cytoplasm uniform, even visible small lipid droplets, sinusoidal clearly visible, hepatic cords neat; model rats lobule ill-defined, swelling of hepatic steatosis, liver cells containing fat droplets of varying sizes, will be pressed against the side of the liver cells, sinusoidal gap narrowed significantly, partial visible fibrous tissue hyperplasia, lobular extend to internal derangement of liver cord; Yiqiyangyin phlegm meridians of Chinese medicine in the treatment groups lobular structure is clear, mild sinusoidal dilatation, structure than normal, and some hepatic steatosis, fat droplets in liver cells was significantly reduced compared with the model group, and many more small lipid droplets exist, hepatic cords arranged in neat. Tips Yiqiyangyin phlegm meridians of Chinese medicine can improve DFL rat hepatic steatosis. 4 of serum TG, TC, HDL levels in serum TG levels showed that: Compared with normal group, model group, low, medium and high dose group was significantly higher serum TG (P lt; 0.05); Compared with model group, serum TG medicine each dose group was significantly lower (P lt; 0.05); middle dose group and high-and low-dose was significantly lower (P lt; 0.05), Chinese high-dose group and low dose No significant between group differences (P gt; 0.05). Levels of serum TC results showed that: Compared with normal group, model group, low, medium and high dose group was significantly higher serum TC (P lt; 0.05); Compared with model group, each group of Chinese medicine significantly lower serum TC (P lt; 0.05); medicine among the three groups, the middle dose group and high-and low-dose was significantly lower (P lt; 0.05), Chinese high-dose group and low dose group had no significant difference between (P gt; 0.05). HDL levels in serum showed that: Compared with normal group, model group, low, medium and high dose group was significantly decreased serum HDL (P lt; 0.05); Compared with model group, the serum medicine HDL was significantly higher (P lt; 0.05); medicine among the three groups, the middle dose group with the high and low dose significantly higher than (P lt; 0.05), Chinese high-dose group and low dose groups was not statistically significant difference (P gt; 0.05). These results suggest that Yiqiyangyin phlegm meridians DFL medicine can significantly reduce serum TG, TC levels, increased serum HDL levels, this may be one of the mechanisms for the treatment DFL. 5 rats in each group liver tissue MDA content and T-SOD activity of MDA in liver tissue showed that: Compared with normal group, model group, low, medium and high dose group was significantly higher (P lt; 0.05 ); Compared with model group, Chinese medicine each dose group was significantly lower (P lt; 0.05); medicine among the three groups, the high dose group and the low and medium dose significantly higher than (P lt; 0.05), low dose between the group and the medium-dose group had no significant difference (P gt; 0.05). The liver tissue T-SOD activity showed that: Compared with normal group, model group, low, medium and high dose group was significantly lower (P lt; 0.05); Compared with model group, Chinese medicine each dose group was significantly increased (P lt; 0.05); medicine comparison between each dose group and high dose group and low and medium dose significantly reduced compared (P lt; 0.05), low dose group and middle dose group had no significant difference between (P gt; 0.05). Tips Yiqiyangyin phlegm meridians of Chinese medicine can significantly reduce the DFL rat liver tissue MDA levels and improved T-SOD activity, significant improvement DFL rat liver antioxidant capacity, this could one of the mechanisms for the treatment DFL. 6 of serum Adiponectin, Adiponectin and AdiR2 liver tissue levels of serum Adiponectin levels, the results showed: Compared with normal group, model group, low, medium and high dose group was significantly lower (P lt; 0.05); Compared with model group, Chinese medicine each dose group was significantly higher (P lt; 0.05); medicine among the three groups, the high dose group and the low and medium dose was significantly lower (P lt; 0.05), low medicine dose group and middle dose groups was not significantly different (P gt; 0.05). Adiponectin levels in rat liver compared to the normal group, model group and Chinese medicine in the treatment groups was not significantly different (P gt; 0.05). The liver tissue AdiR2 level, the results showed: Compared with normal group, model group, low, medium and high dose group was significantly lower (P lt; 0.05); Compared with model group, Chinese medicine each dose group was significantly increased (P lt; 0.05); medicine among the three groups, the higher dose group and low dose significantly increased (P lt; 0.05), high-dose group and TCM low dose group had no significant difference between (P gt ; 0.05). These results indicate that Yiqiyangyin phlegm meridians DFL medicine can significantly improve the level of serum and liver AdiR2 Adiponectin levels, but no significant effect of liver Adiponectin levels, suggesting that this side effect may be related to the treatment of DFL DFL large increase Rat Serum Adiponectin levels and liver AdiR2 level, whereas its effect on the liver Adiponectin levels irrelevant. Conclusions: 1 Yiqiyangyin phlegm meridians of Chinese medicine can significantly improve DFL rat hepatic steatosis, reduce liver index, for DFL has a good therapeutic effect. 2 Yiqiyangyin phlegm meridians of Chinese medicine can significantly reduce the DFL rat serum TG and TC, elevated serum HDL levels, suggesting that the DFL therapeutic effect and regulate blood lipids related. 3 Yiqiyangyin phlegm meridians of Chinese medicine can significantly improve the DFL rat liver antioxidant capacity and delay \fibrosis, this may be the traditional Chinese medicine treatment of DFL one of the mechanisms. 4 Yiqiyangyin phlegm meridians DFL medicine can be significantly increased levels of serum and liver AdiR2 Adiponectin levels, suggesting that this effect may contribute to the role of traditional Chinese medicine treatment of DFL one of the mechanisms, and their effects on the liver Adiponectin levels irrelevant.
