Abstract


Multiple sclerosis (MS) is a chronic inflammatory demyelinating disease of the central nervous system, occurs in young adults, recurrent, delayed healing, is recognized worldwide as one of the difficult disease. Because of MS pathogenesis is not clear, so at this stage is still no sure and effective prevention and treatment measures. Therefore has important theoretical significance and clinical value of the study and discussion of the etiology, pathogenesis of MS, looking for effective prevention and control measures. Experimental autoimmune encephalomyelitis (Experimental autoimmune encephalomyelitis, EAE) is sensitive experimental animal central nervous system (Central nervous system, CNS) autoimmune disease, animal models of CNS inflammatory demyelinating disease of the classic . In this study, by to establish EAE models, testing the mechanism of action of acupuncture to alleviate period of of EAE rat IL-23 and sICAM-1 impact of to explore acupuncture treatment EAE, comparing acupuncture treatment efficacy of hormone therapy EAE. Methods: 49 female Wistar rats were randomly divided into blank control group, EAE model group, electro-acupuncture group, and hormone therapy group. Fetch the guinea pig spinal cord homogenate with complete Freund's adjuvant mix made the oil emulsion immune induced, and intraperitoneal injection of Bordetella pertussis vaccine within 72h secondary immunization, preparation of EAE model. Blank control group footpad limbs of saline injected rats, intraperitoneal injection of saline within 72h. Blank control group and the the EAE model group was not any treatment. Electro-acupuncture group acupuncture Baihui bilateral Shenshu Zusanli, the ipsilateral shenshu Zusanli pick a set of positive and negative. Hormone therapy group was first given intravenous injection of methylprednisolone sodium acetate, and then orally prednisone acetate suspension. From the first 10d after modeling, daily observation of the general condition of the rats were recorded, changes in body weight and nerve function scores in the experimental section 30d (in remission) drawn by enzyme-linked immunosorbent assay (Elisa) serum IL-23 and sICAM-1 levels. Results: (1) by guinea pig spinal cord homogenate and complete Freund's adjuvant mix made emulsion immune Wistar female rats successfully made experimental autoimmune encephalomyelitis (EAE) model. (2) The neurological score: peak period (after modeling 18d) hormone therapy group and electric needle treatment group and model group neurological score was extremely significant with difference (P lt; 0.01); remission period (after modeling 30d) compared with the model group, the group of hormone therapy, electro-acupuncture group neurological score was a significant difference (P lt; 0.05); hormone therapy group and electro-acupuncture group was no significant difference (P gt; 0.05). (3) IL-23 of the detection results: of EAE model group and blank control group comparison there is a very significant with difference (P lt; 0.01), electro-acupuncture group, hormone therapy group and blank control group showed no significant forward difference (P gt; 0.05), electro-acupuncture group EAE model between the groups was significant difference (P lt; 0.05), a highly significant difference (P lt; 0.01) between the hormone therapy group with EAE model group, electro-acupuncture treatment between the group and the hormone therapy group no significant difference (P gt; 0.05). (4) levels of sICAM-1 the detection results: of EAE model group and blank control group comparison there is a very significant with difference (P lt; 0.01), electric needle the treatment group, hormone therapy group and the control group showed no significant difference (P = gt; 0.05), hormone therapy group and the model group between a significant difference (P lt; 0.05), electro-acupuncture group with EAE the model group significant difference (P lt; 0.05), but the electro-acupuncture group between the hormone therapy group was no significant difference (P gt; 0.05). Conclusion: Acupuncture therapy and hormone therapy can alleviate EAE inflammatory response by inhibiting IL-23, sICAM-1 expression; acupuncture therapy and hormone therapy can make the remission of IL-23, sICAM-1 remains relatively normal The level; acupuncture therapy and hormone therapy EAE has the same effect.
