The Detection and Clinical Implication of Methylation of RAR-β Gene in Patients with Non-small Lung Cancer

Abstract


Objective: To study non-small cell lung cancer patients the cancerous tissue RAR - beta gene methylation and its clinical significance , to explore the pathogenesis of lung cancer , early diagnosis and prevention provide a theoretical basis . Methods: 80 patients with non- small cell lung cancer , lung cancer and adjacent normal tissue RAR - beta gene methylation detected by methylation-specific PCR ( MSP ) . Results: cancer tissue of patients with lung cancer , the cancer tissue extracted genomic DNA as a template for MSP amplification . PCR showed that 80 cases of lung cancer and corresponding adjacent tissue samples simultaneous detection of 46 cases of lung cancer tissue samples methylation positive fragments adjacent tissue samples in 14 cases of methylation positive fragments , methylation rates were 57.5 % and 17.5% , two sets of methylation rate comparison , the difference was statistically significant ( P lt ; 0.01 ) ; clinical stage 0 ~ Phase I and II to Phase IV and central and peripheral compared to RAR - beta gene methylation rate difference was not statistically significant ( P gt ; 0.05 ) ; different pathological type of lung cancer tissue RAR - beta gene methylation test results showed that the detection rate of squamous cell carcinoma of RAR - beta gene methylation adenocarcinoma and other , the difference was statistically significant ( P lt ; 0.05 ) Conclusion : MSP is a simple detection of the methylation status of CpG -rich region , a sensitive and specific method ; lung cancer in the presence of RAR - beta gene promoter CpG island methylation of promoter CpG island methylation and lung cancer development closely related to the detection of RAR - beta gene methylation contribute to the diagnosis of lung cancer ; RAR - beta gene methylation rate has nothing to do with age, sex , clinical stage and clinical typing and other clinical factors in lung cancer may be associated with pathological type , smoking.
