Abstract


The purpose of bladder transitional cell carcinoma of the urinary tract is the most common malignancy in the incidence of malignant tumors in the world fifth ranked second in the urinary tract diseases mortality. The trend of increased incidence in recent years, has a multi-center, easy to relapse, and invasive growth characteristics, and use more comprehensive treatment of surgery, the treatment is still not ideal. Urgent need for effective treatment. In recent years, the rapid development of molecular biology, provides a theoretical basis for looking for new and effective treatment of transitional cell carcinoma of the bladder. Studies have shown that the development of bladder transitional cell carcinoma is a polygenic multifactorial comprehensive results, including cell cycle regulation plays an important role. Polo like kinase 1 (polo like kinase 1, PLK1) is widely present in eukaryotic cells was the expression of cyclin-dependent serine / threonine kinase, play an important role in mitosis and cell proliferation is closely related to, and some of the biological behavior of a variety of tumors and poor prognosis. This study investigated the bladder transitional cell carcinoma of PLK1 Expression and clinical significance of Cyclin B1, may provide a theoretical basis for the early diagnosis and treatment of bladder transitional cell carcinoma of the. Collected 41 cases of bladder transitional cell carcinoma of the surgical resection specimens and 10 cases of normal bladder tissue (from the tumor more than 2cm cancer adjacent normal mucosa), all specimens were fixed in formalin solution, all organizations were confirmed by pathology using immunohistochemistry SP method in the detection of both of PLK1 positive expression of Cyclin B1 analysis both with clinical and pathological parameters of contact between the correlation. The results of bladder transitional cell carcinoma of PLK1 expressed mainly in the nucleus was brown or tan, some pulp, nuclear simultaneous expression, Cyclin B1 is expressed mainly in the cytoplasm, is brown or tan. Of PLK1 in bladder transitional cell carcinoma, the positive expression rate of Cyclin B1 were 51.2.0% (21/41), 56.1% (23/41), normal bladder tissue PLK1 and Cyclin B1 protein was weakly positive or no expression The difference was statistically significant (P lt; 0.01). Independent of PLK1 expression with age, sex, clinical stage and lymph node status (P gt; 0.05), with the pathological grade, clinical stage, pathological grade differentially expressed with statistical significance (P lt; 0.05 ), the positive expression rate of 75.0%, 22.2%. Independent of Cyclin B1 expression with age, gender, lymph node status (P gt; 0.05), clinical stage and pathological grade is closely related with the clinical stage increased elevated positive expression rates were 40.0%, 71.4 % difference was statistically significant (P lt; 0.05) in different clinical stages, and increased with increasing pathological grade, the difference was statistically significant (P lt; 0.05). PLK1 and Cyclin B1 there is a certain degree of correlation (r = 0.317) between the expression in bladder transitional cell carcinoma. Conclusion PLK1 and Cyclin B1 in bladder transitional cell carcinoma of high expression and pathological grade, clinical stage is closely related to the prompt of PLK1 and Cyclin B1 play an important role in the onset and progression of bladder transitional cell carcinoma, and there existed a certain regulating relations, PLK1 and Cyclin B1 is expected to provide a new theoretical basis and a new therapeutic target for the treatment of bladder transitional cell cancer on the molecular level research.
