Effects and Mechanism of Hepatoma-derived Growth Factor in Cell Growth, Migration, and Invasion of Breast Cancer and Prostate Cancer Cells

Abstract


Objective: To assess the role of HDGF in the migration of and invasion by prostate and breast cancer cells in vitro.Methods: siRNA-mediated HDGF knockdown was achieved in DU145 and MCF7 cells, and the effect of such knockdown was assessed by using MTS, [3H]-thymidine incorporation and Transwell assays. Moreover, using PCR array, we identified differentially expressed genes that might mediate the HDGF-induced cellular effects.Results: Down-regulation of HDGF expression significantly reduced the proliferation of both DU145 and MCF7 cells. However, down-regulation of HDGF expression in DU145 inhibited cell migration and invasion, but in MCF7 cells it stimulated cell migration and invasion. Real time PCR shows that downregulation of HDGF lead to upregulation of gene PIK3R1 in MCF7, and upregulation of gene SERPINE1 in DU145.Conclusion: HDGF may participate in the pathogenesis of prostate and breast cancers by promoting cell growth, in the DU145 by promoting cell migratrion and invasion, in the MCF7 by inhibiting cell migratrion and invasion. HDGF may participate in cell migration and invasion by differential pathway, however, it need to further study .
