Part 1 The impact of over expression of connective tissue growth factor(CTGF) on human pancreatic cancer cellsObjective:To explore the impact of connective tissue growth factor (CTGF) on the cell growth、proliferation、invasion and migration of human pancreatic cancer PANC-1 cells.Methods:The MTT assay and colony-forming unit assay were employed to test the ability of PANC-1 cell growth and proliferation after recombinant adenovirus mediated CTGF over-expression ; And that migration and invasion capacity were observed by transwell assay in human pancreatic cancer PANC-1 cells.Results:High expression of CTGF suppressed the cell growth and proliferation of PANC-1 cells by colony-forming unit assay and MTT assay. The ability of migration and invasion were suppressed as well when CTGF over expressed by transwell assay.Conclusions:Over expression of connective tissue growth factor (CTGF) might inhibit the cell growth、proliferation、invasion and migration of human pancreatic cancer cells. Part 2 The impact of connective tissue growth factor(CTGF) on the cell cycle and the related genes of human pancreatic cancerObjective:To explore the impact of connective tissue growth factor (CTGF) on the cell cycle of human pancreatic cancer PANC-1 cells and the role of its related genes.Methods:Cell cycle changing was tested after recombinant adenovirus mediated CTGF over-expression by flow cytometry. To explore the impact of PANC-1 cells express cyclin D1 protein by western-blot.Results:High expression of CTGF suppressed the PANC-1 cells in the G1 Phase of the cell cycle, Inhabit cells copy and enter mitosis by flow cytometry. Protein expression in pancreatic cancer cells by Western bolt: High CTGF expression after PANC-1 cells,Cyclin D1 protein expression was significantly reduced.Conclusions:High expression of CTGF might reduce cyclin D1 protein expression in pancreatic cancer cells, led to PANC-1 cells arrest in the G1 Phase of the cell cycle, and to affect the proliferation、invasion and migration of human pancreatic cancer cells.
