Abstract


Objective: In this study through researching the invasive gene expression changes、the number of cancer cell which were throughed the basement membrane and growth activity in vitro of hepG2 liver cancer vaccines which were treatmened by 60Co,we want to verify initially the invasion and growth activity changes of liver cancer vaccine.Methods: Culturing three hepatoma cells in vitro (A.wild-type HepG2 hepatoma cells ;B.transfected with hGM-CSF gene in HepG2 hepatoma cells; C.60Co irradiation of the hGM-CSF gene transfected HepG2 liver cancer vaccine), three were detected by MTT kinds of cells in 24h, 48h, 72h of the OD values and plot the growth curve; Using in vitro transwell chamber invasion assay to observe the three cells under the same conditions and the same time through the small room of the basement membrane of their cell number; through RT-PCR detecting expression of the three rinds cells’matrix metalloproteinase-2 （MMP-2） at the mRNA level changes;Results: The OD values of the hGM-CSF gene transfected HepG2 liver cancer vaccine group which were irradiated by 60Co under the same culture conditions, the same point in time significantly smaller than the other two groups and the difference was significant （P <0.05）. Three types of cells （A、B、C cell types above） transwell invasion test showed:③group the number of cells through artificial basement membrane was significantly less than the first two groups; PT-PCR showed: liver cancer vaccine group of cells in the MMP-2 mRNA was significantly lower than the other.Conclusion: The invasion and growth activity of hGM-CSF gene transfected HepG2 liver cancer vaccines which were processed by 60Co was significantly reduced.
