Abstract


Objective: To investigate small doses of radiation on human renal clear cell carcinoma 786-0 cell immunogenicity calreticulin (CRT), MHC-I and MHC-II expression amount, and to explore the impact of radiotherapy on the immunogenicity of the tumor cells, , to further improve the efficacy of cancer treatment to provide basic experimental data for better play synergistic anti-tumor effect of radiotherapy and immunotherapy. Methods: CCK-8 assay radiotherapy the 786-0 cell growth inhibition; different doses of radiation acting on 786-0 cells after 24 h by flow cytometry cell apoptosis was detected by indirect immunofluorescence and membrane surface of the CRT , expression of MHC-I and MHC-II molecules. Results: 1 CCK-8 test results: 0Gy, 6Gy, 12 Gy, 24 Gy four groups of irradiation on the 786-0 cell proliferation inhibition rate with increasing dose increased. The half inhibitory dose (IC50): 12Gy. 2 0Gy 6Gy, 12 Gy, 24 GY four groups irradiation on the the 786-0 cell surface expression of CRT: four different dose intervention of 786-0 cells after 24 hours, flow cytometry cell surface expression of CRT in the amount of , corresponding to the mean fluorescence intensity were: control group: CRT (0Gy) 13.32% ± 2.01%; treatment groups: the CRT (6Gy) 78.25% ± 5.83%; CRT (12Gy) 202.24% ± 16.47%; CRT (24Gy) 83.51 % ± 8.26%. Each group were compared with the control group twenty-two, there were significant differences (p lt; 0.001). To 12Gy group for the strongest, followed by 24Gy weakest at 6Gy group. With the increase of the dose, the mean fluorescence intensity of calreticulin first gradually increased, after a downward trend.
