Objective : To study the uremic patients with RORγt and Foxp3 gene expression , clearly Treg, Th17 two kinds of cells in uremic patients balance changes, changes in uremic patients explore this micro- inflammatory state 's role and right hemodialysis patients influence the development of cardiovascular disease methods: 30 patients with uremia , which did not enter the 10 patients on dialysis (CKD group ) , 20 patients entering dialysis (MHD group ) , will MHD groups according to cardiac function were divided into two groups, LVEF gt; 50% of MHDl group , LVEF lt; 50% of MHD2 groups. Select both cardiac function and renal function is normal healthy control group of 10 patients (A group ) . Fasting experimental group were pumping blood with EDTA anticoagulant tubes 2m1, and then apply the extracted cell lymphocyte separation medium , serum alternate upper layer was collected , and RNA was extracted from lymphocytes further conducted after RT-PCR, semi quantitative analysis of the transcription factor Foxp3 Treg cells and Th17 cells transcription factor RORγt, for further analysis , Treg and Th17 balance of the two cell change . By ELISA IL-6, IL-10, IL-17 levels . And then the transcription factor Foxp3 transcription factor RORγt and IL-6, IL-10, IL-17 correlation analysis . Results: The completely randomized design analysis of variance , compared to the normal control group and uremic patients and found Foxp3 and RORγt expression was significantly unbalanced (P lt; 0.05), in the uremic group RORγt expression was significantly increased, while Foxp3 were significantly decreased in MHD2 group this imbalance was more serious (P lt; 0.01), but in the CKD group and MHD1 groups did not differ (P gt; 0.05), through correlation analysis, we found RORYt and IL-6, IL- 17, hsCRP was positively correlated with IL-10 is negatively correlated ; Foxp3 and IL-10 was positively correlated with IL-6, IL-17, hsCRP negative correlation . CONCLUSION : Treg and Th17 cells in the presence of uremic patients imbalance , and this imbalance is not only state of uremic patients microinflammatory , but also in maintenance hemodialysis patients in the development of cardiovascular disease plays a synergistic effect .
