Abstract


Purpose by measuring the in vitro culture of normal SD rats and the chronic prostatitis SD rat prostate smooth muscle cells intracellular calcium ion concentration to understand chronic prostatitis prostate smooth muscle cells and smooth muscle cells of the normal prostate free calcium ion concentration differences and their significance, to explore the role of intracellular free calcium ions in the development of chronic prostatitis occur. Methods 50 rats divided into inflammatory group and the normal control group, 10 of normal SD rat prostate protein extraction and purification, respectively, in the first 0,30 d inflammation group each rat Pinedo point injection of rat prostate protein purification solution and FCA emulsion (1:1 suspension) 1.0 ml, a concentration of 40mg/ml, 0. 5 ml each intraperitoneal injection of DPT vaccine to cause autoimmune chronic prostatitis, the other to take the same method of normal SD rats injected with saline as a control group; modeling after 45 days, each rat to take part of the prostate tissue, paraffin-embedded sections of the HE staining histological observation, confirmed the successful model in vitro SD rats were cultured prostatic smooth muscle cells, the use of differential adhesion the purified prostate smooth muscle cells, smooth muscle cells by immunohistochemistry SP method was confirmed to cultivate cells cells from a cell suspension; digestion, highly specific calcium ion fluorescence indicator Fluo 3-AM was added to the cell suspension, set carbon dioxide incubator and incubated for 1h load cells, centrifuged to remove excess dye. Calcium-free buffer repeatedly washed three times, the last application of flow cytometry to detect the concentration of intracellular calcium. Fluo 3-AM, detected with a wavelength of 480nm excitation SD rats of chronic prostatitis group and control group the SD rat prostate smooth muscle intracellular calcium ions in the emission wavelength of the the 525nm fluorescence intensity value analysis comparing the measured results by flow cytometry parallel fluorescence intensity fluorescence value of the model group was significantly higher than the normal control group, SD rats (P lt; 0.05), the difference was significant. Conclusion When using a 480nm wavelength excitation Fluo 3-AM, the fluorescence intensity of free calcium ions is proportional to the concentration of the above results show that the intracellular calcium concentration in chronic prostatitis SD rat smooth muscle cells than normal SD rats were elevated, we believe that prostatic smooth muscle cells of chronic prostatitis caused by intracellular free calcium concentration increased, resulting in calcium homeostasis fluctuations, intracellular calcium overload in a series of activities, thereby affecting the physiological function of the prostate tissue and cell intracellular free calcium concentration in the prostate smooth muscle cell changes may be an important factor for the incidence of chronic prostatitis, PSMCs intracellular free calcium concentration may play a role in the pathogenesis of.
